| Objective:To observe expression of P-p38MAPK and content change of collagen type â…¡ in the nucleus pulposus tissue of degenerative intervertebral disc, and to study the relationship between P-p38MAPK and the severity of degeneration of the intervertebral disc.Content:The HE staining and observed pathological differences of the experimental group and the control group of the nucleus pulposus tissue under light microscope;Immunohistochemical method for the collagen type â…¡ content the experimental group and the control group of the nucleus pulposus tissue; Immunohistochemical method for the P-p38MAPK expression of the experimental group and the control group in the nucleus pulposus cells.Methods:Selected40lumbar intervertebral disc specimens of operation patients. Divided into2groups. First group (experimental group):25cases from patients with lumbar disc herniation. There were male17, female8, age32-65years old. The second group (control group):a total of15cases, male11, female4, age6-36years old.12of them from the young patients of lumbar trauma,2from the lumbar vertebral tuberculosis patients;1from a child of the fifth lumbar hemivertebra deformity patient. All the patients had no back pain history, and the examination of MRI and CT had no protrusion of lumbar intervertebral disc.All the specimens were produced for the wax block and frozen section. HE staining and observed pathological differences of the experimental group and the control group of the nucleus pulposus tissue under lightmicroscope.P-p38MAPK expression was determined by the immunohistochemical method. Judgment of positive result:the expression of P-p38MAPK protein in the cell nucleus, yellow or brown granules. Collagen type â…¡ expression was determined by the immunohistochemical method.Judgment of positive result:collagen type â…¡ expression in the matrix and cytoplasm in expression, presenting buffy.Applying image analysis software to evaluate the expression of P-p38MAPK and collagen type â…¡ in the corresponding region by immunohistochemical staining,and automatically calculate the gray value to view expression intensity. Gray grade0-255,level0is the deepest, level255is the most shallow.The smaller the value is, the stronger the expression is. The field average gray is the result of determination. Each specimen was selected7views, then the average was taken as the average gray value of the specimen. The data of the experimental group and the control group was used for statistical analysis by double free samples T test. The final analysis was the correlation between P-p38MAPK and collagen type â…¡ in each specimen.Result:HE staining was observed under ordinary eyepiece:control group:in nucleus pulposus tissue slices nucleus pulposus cells distributed in the extracellular matrix, and they were round or oval with a large number.The orderly arrangement of collagen around the cells formed a mesh structure.Experiment group:nucleus pulposus cells and chondrocytes were fewerã€larger and irregular shape," appeared "vacuole sign" in the cytoplasm.The arrangement of collagen fibres around the cells was chaos with mucinous variant.There were visible voids or fracture of the fibrous structure.P-p38MAPK results of Immunohistochemical determination are as follows:in the control group, P-p38MAPK expression is less, average gray value of198.81±17.52; the experimental group P-p38MAPK expression is more, the average gray value of115.99±21.11; the difference is significant(P<0.05).Immunohistochemical determination of collagen type â…¡ results are as follows:in the control group,collagen type â…¡ expression is more, the average gray value of163.52±11.27;the experimental group of collagen type â…¡ expression is less, average gray value of187.98±17.25; the difference is significant(P<0.05).With the degeneration of intervertebral disc in nucleus pulposus cells P-p38MAPK expression gradually grows in quantity,and collagen type â…¡ content gradually decreases.There is a relationship between the two. The correlation coefficient is r=-0.731.Conclusion:1The expression of P-p38MAPK lies in the degenerative lumbar disc tissue.2With the aggravating of degenerative degree in the lumbar disc tissue, P-p38MAPK expression amount will increase gradually.P-p38MAPK participates in the disc degeneration process, and it is one of the reasons for lumbar intervertebral disc degeneration. |
