[Objective] To analysis Thalidomide’s effection on the expression of VEGF and MVD and pathogenesy in immune-mediated aplastic anemia mouse model group by comparison with that of control group,and establish the theoretical basis for clinical treatment.[Methods] Immunohistochemical assay was performed to test the the expression of VEGF and MVD of pathological sections of bone marrow of immune-mediated aplastic anemia mouse model group and control group. VEGF’s positive site is located at cytoplasm. The positive cells are brown.The image was observed through light microscope. The results of immunohistochemical staining is converted into the mean optical density through the computer analysis system.[Results] The mean optical density of VEGF for immune-mediated aplastic anemia mouse model group was0.186±0.005. the positive site is located at cytoplasm. The mean optical density of the thalidomide intervention AA group was0.190±0.001. the expression of VEGF in immune-mediated aplastic anemia mouse model group was no significantly lower than the thalidomide intervention AA group (P>0.05). The mean optical density of MVD for immune-mediated aplastic anemia mouse model group was0.286±0.005. The positive site is located at cytoplasm. The mean optical density of MVD for the thalidomide intervention AA group was0.286±0.005. the expression of MVD in immune-mediated aplastic anemia mouse model group was no significantly lower than control group (P>0.05).[Conclusion] the expression of VEGF in immune-mediated aplastic anemia mouse model group was significantly lower than normal group and would be descending under the circumstance that eshematopoietic failure.the expression of MVD in immune-mediated aplastic anemia mouse model group was significantly lower than normal group and was closely correlated with the injury of hematopoietic microenvironment. The decrease of VEGF and MVD presumably lead to nutritional disturbance for hematopoietic microenvironment and the failure of hematopoietic stem cell. Compared with the AA group,the thalidomide intervention AA group was no significant difference. In this experiment,the thalidomide was no significant effection for the treatment of AA.
