Obejective:To explore the effects of Jin Lida Granule on insulin resistance(IR) in high-fat diet fed SD rats and its possible mechanisms.Methods:45SD male rats(140-160g) were induced to be insulin resistance by fed with high-fat diet for12weeks and then were divided into four groups randomly:High-fat diet group(HF group),Jin Lida Granule group,Metformin group,and the intergration of Jin Lida Granule with Metformin group. Paired normal-diet group was also matched.During the treatment,fasting and postchallenge blood glucose levels were detected by IPGTT at week4,8,and10,and and area under the curve of blood glucose(AUC of blood glucose)was calculated.Serum levels of insulin were tested at week10and HOMA-IR,ISI were also calculated.After ten weeks’ treatment,the rats were killed and blood serum was collected to detect TG,Tch,HDL-C,LDL-C,ALT,AST,TBIL, DBIL,TNF-a,IL-6,FFA,SAA,Leptin,Adiponectin,and hepatic histology was analyzed by HE and oil red O staining.Results:In the aspect of glucose tolerance and serum insulin level,compared with HF group,both Jin Lida Granule group and Intergrated group had a lower blood glucose.smaller AUC of blood glucose at week eight and ten(p<0.05)and lower serum insulin at week ten(p<0.05).At the same time,both of Jin Lida Granule group and Intergrated group had lower HOMA-IR and higher ISI than HF group.In aspect of lipids, Intergrated group and Jin Lida Granule group had lower serum TG level than HF group(p<0.05),and different decreased extent of Tch,HDL-C and LDL-C.In aspect of liver function,both of Jin Lida Granule group and Intergrated group had a lower serum level of ALT,ALT,TBIL and TBIL than HF group, and had a significant difference on ALT and AST(p<0.05).In aspect of serum level of adipocytokines,compared with HF group,Jin Lida Granule group and combined group had notable lower level of Leptin,TNF-a,IL-6.higher level of Adiponectin(P<0.05),and decreased trend of SAA.On the serum level of FFA,there is no decreased trend of Jin Lida Granule group,but combined group had a significant decrease(P<0.05). HE and oil red O staining of liver indicated that liver cells of HF group had a severe fatty change,and both of Jin Lida Granule group and Intergrated group had significant lower steatosis of liver than HF group.Conclusion:Firstly.Jin Lida Granule group can correct abnormal glucose tolerance of IR rats,decrease level of insulin,improve the insulin sensitivity.these phenomenon indicated that Jin Lida Granule could improve IR though regulating glucose metabolism,and the specific mechanism still be needed further study.Secondly, Jin Lida Granule can decrease the serum level of TNF-a,IL-6,SAA,Leptin,and raise level of Adiponectin,these mean Jin Lida Granule can improve IR by inhibiting the imflmmatory function and promoting serum level of Adiponectin. Thirdly, Jin Lida Granule can decrease the serum level of TG,Tch,HDL-C and LDL-C,these phenomenon showed that Jin Lida Granule can improve IR by correcting lipid metabolism disorder.At last, Jin Lida Granule can improve liver function and decrease the extent of liver steatosis significantly, these phenomenon indicated that Jin Lida Granule could decrease the accumulation of lipids in liver.
