Baoligan capsule is functional food that against chemical liver injury. It is composed of several Chinese herbal medicines:Curcuma longa L, Schisandra chinensis baill, Alisma orientalis (Sam.)Juzep., Paeonia lactiflora Pall, and Rosaceae. This capsule can protect liver cells from free radicals and other physical chemical damage, coordination of metabolism to maintain normal function of liver cells. This paper studied on Quality Standard of Baoligan Capsule, in vitro antioxidant activity, Protective Effect of Baoligan Capsules on Non-alcoholic Fatty Liver Diseases in Rats and Stability of Curcumin in the Production Process of Baoligan Capsule.First, The quality standard for Baoligan Capsule was to been established. Curcuma longa, Schisandra chinensis, Alisma orientalis, Paeonia lactiflora and Mespilus germanica were identified by TLC, and the contents of curcumin and schisandrin were determined by RP-HPLC. The TLC spots were clear and the resolution was good while the content of curcumin was determined by RP-HPLC. Curcumin showed good linear relationship at a range of5.15-102.90μg/mL, r=0.9999. The average recovery of curcumin was98.04%(n=9) and RSD was3.15%by RP-HPLC. And schisandrin showed good linear relationship at a range of5.22-104.40μg/mL, r=0.9998. The average recovery of schisandrin was98.52%(n=9) and RSD was3.67%by RP-HPLC. The method is specific, reliable and accurate. It can be used for the quality control of Baoligan Capsule.Second, Study on Thermal Stability of Curcumin in the Production Process of Baoligan Capsule, to optimize operating conditions. According to the process conditions, curcumin was heated at different prescription and solvent, then detected its concentration which was changed with time by HPLC. Establish the degradation rate equation of Curcumin by kinetic method, and calculate its half-life. White Peony Root, Fructus Schisandrae and Alisma orieniale extract solution can accelerate the degradation of curcumin respectively, and Alisma orientale has the most influence on the degradation of curcumin. Extract of curcumin degradation rate decreased with increasing ethanol concentration and reach the maximum in the water. Curcum should be extracted solely during extract process, shorten heating time as much as possible, the effect of spray drying.is better than the effect of vacuum drying.Thirdly, in vitro antioxidant activity of Baoligan capsules has been investigated. The study on the DPPH scavenging ability of theaflavins, O2-. scavenging ability,.OH scavenging capacity, reducing power and antioxidant activity to liposomes show that Baoligan capsules has strong antioxidant activity in vitro, Which inhibit antioxidant activity of liposomes are obviously stronger than the reference substance VE.At last, to study on the effect of Baoligan capsules on the non-alcoholic fatty liver disease (NAFLD) in rats induced by the lack of methionine choline diet (MCD). The model of NAFLD was established by MCD, and the rats were administered Baoligan Capsules (300and600mg? kg-1) for40days. Liver samples were fixed in formaldehyde and paraffin-embedded. The steatosis, inflammation and fibrosis of liver were assessed after HE staining, and the content of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) in plasma, and triglyceride (TG), total cholesterol (TC), malondialdehyde (MDA), nitric oxide (NO) in liver and liver index were determined. Baoligan capsules could protect against hepatic steatosis and inflammation, Reduce the liver index, ALT, AST, MDA, NO, TG, TC in liver significantly. Baoligan capsule has a protective effect on NAFLD in rats induced by MCD.Above all, Baoligan capsule has improved quality standards, scientific production technology, antioxidant activity in vitro and efficacy significantly in vivo, it can be further researched and developed.
