Part1Killing effect of BTW5on adult and juvenile Schistosoma japonicum invitroObjective: To observe the killing effect of BTW5on both juvenile and adultworms of Schistosoma japonicum in vitro.Methods: The mice were infected with the cercariae of Schistosoma japonicumobtained from infected Oncomelania.5weeks later, adult worms were obtained fromthe infected mice by perfusion of the livers and mesenteric veins. Juvenile worms wereobtained after18days infection. The adult and juvenile worms were cultivated inDMEM medium containing different concentrations of BTW5and were observed for3days. After the observation, the worms were stained by hydrochloric acid carmine toobserve the injuries.Result: The death rates and motility reducing rates of adult and juvenile worms inDMEM medium containing different concentrations of BTW5were significantly higherthan the controls. The staining suggested that BTW5had a damage effect on thetegument and the intestinal canal of the male and female worms, including the ovariesof the female worms.Conclusion: BTW5had an antischistosomal effect on adult and juvenile worms.Part2Activity of BTW5against Schistosoma japonicum in vivoObjective: To observe the killing effect of BTW5on Schistosoma japonicum invivo.Methods: Mice were infected with40cercariae. These infected mice were dividedinto three groups. The mice in first group were given BTW5by tail vein injection after9d infection. The mice in second group were given the drug of after24d infection. Themice in third group were given the drug after35d infection. The three groups were allgiven the drug of three does of50,100and200mg/kg, continued5days. The mice in PZQ group were given PZQ of the does of300mg/kg, continued3days. After7weeks,the mice were dissected and collected the worms. Calculate the worm reduction rate, thefemale worm reduction and the egg reduction rate. The liver tissue sections wereexamined under a light microscope to investigate the changes of the granuloma causedby BTW5.Result: The number of worms of BTW5experimental groups were all significantlylower than the controls(P<0.05),and the female worms of BTW5experimental groupswere all significantly lower than the controls(P<0.05). Under the microscope, the liverslices in the treatment group shows fewer granulomas, and the liver cells withoutobvious pathological changes arranged in neat rows.Conclusion: BTW5have a killing effect on the worms of3developmental stages invivo. Especially, the35d old groups were the most susceptive to BTW5of thehighest-dose of200mg/kg.Part3Activity of5extract of Pulsatilla chinensis against Schistosomajaponicum in vivoObjective: To observe the killing effect of5extract of Pulsatilla chinensis onSchistosoma japonicum in vivo.Methods: Mice were infected with40cercariae. These infected mice were dividedinto three groups. The mice in first group were given the drugs by tail vein injectionafter5d infection. The mice in second group were given the drugs of after20d infection.The mice in third group were given the drugs after35d infection. The three groups wereall given the drugs of the dose of200mg/kg, continued5days. The mice in PZQ groupwere given PZQ of the does of300mg/kg, continued3days. After7weeks, the micewere dissected and collected the worms. Calculate the worm reduction rate and thefemale worm reduction.Result: In the5d group, the number of worms of experimental groups were allsignificantly lower than the control(sP<0.05). The number of female worms of I, II andV experimental groups were lower than the controls(P<0.05). In the20d group, thenumber of worms of I and III experimental groups were all significantly lower than thecontrols(P<0.05). The number of female worms of I experimental groups were lowerthan the controls(P<0.05). In the35d group, the number of worms of I, II, III and Vexperimental groups were all significantly lower than the controls(P<0.05). The number of female worms of all experimental groups were lower than the controls(P<0.05).Conclusion: Extract V had the best killing effect of the5extract on35d age adultworms and female worms in vivo.
