Objective:To investigate the damage and repair of high doses of X-ray on human peripheralblood nuclear cell DNA by single cell gel electrophoresis, SCGE. To investigate thedamage of semen by different doses of X-ray in vitro. To detect the DNA damage ofdifferent doses of X-ray on human peripheral blood nuclear cell and sperm by singlecell gel electrophoresis, SCGE. Evaluation the level of DNA damage of humanperipheral blood nuclear cell and sperm after different doses of X-ray.Methods:1.using human peripheral blood with normal blood routine, give the dose of0Gy,2Gy,4Gy,6Gy,8Gy,10Gy X-ray radiation with energy of6MV. Detect thepercentage of comet-like tail, tail length and content of DNA in tail of whole bloodcell’s DNA by single-celled gel electrophoresis technology in0.5h,1h,2h.2.using human semen with normal seminal fluid routine, give the dose of0Gy,2Gy,4Gy,6Gy,8Gy,10Gy X-ray radiation with energy of6MeV in vitro. Detectthe seminal fluid routine mainly focus on sperm motility rate and sperm viability, thepercentage of comet-like tail, tail length and content of DNA in tail of sperm DNAby single-celled gel electrophoresis technology in1h.3. using human peripheral blood with normal blood routine and normal sperm, givethe dose of0Gy,2Gy,4Gy,6Gy,8Gy,10Gy X-ray radiation with energy of6MU.Detect the percentage of comet-like tail, tail length and content of DNA in tail of wholeblood cell’s DNA and sperm’s DNA by single-celled gel electrophoresis technology in1hour. blood cell’s DNA and sperm’s DNA by single-celled gel electrophoresis technology in1hour.Results:1. The comet rate is1%in0Gy group,27.10%in2Gy grup,76.32%in4Gy group, above90%,nearly100%in6Gy,8Gy,10Gy.The comet rate has reduced experience2h repair in2Gy,4Gy,have statistical significance; the comet rate has no significantly reduce in6Gy,8Gy,10Gy,have no-statistical significance.DNA repair fast in1h,slower in1h-2h.2. there is no-statistical significantly differences between experiment groups and control group in sperm motility rate and sperm viability. The comet rate is47.37±8.32%,92.22±9.025%,100%,100%,100%in2Gy,4Gy,6Gy,8Gy,10Gy group respectly,is significantly high in contrast with control group,have statistical differences.3. The peripheral blood nuclear cell’s and sperm’s comet rate is1.00±0.10%,2.1±1.5%, respectively, have a evidently variance in0Gy group(υ=18, t=2.31>1.734,p<0.05). The peripheral blood nuclear cell’s and sperm’s comet rate is27.10±4.71%,47.37±8.32%, respectively, have a evidently variance in2Gy group(υ=18, t=6.76>1.734,p<0.05). The peripheral blood nuclear cell’s and sperm’s comet rate is76.32±10.28%,92.22±9.02%, respectively, have a evidently variance in4Gy group(υ=18, t=3.68<1.734,p<0.05). The peripheral blood nuclear cell’s and sperm’s comet rate is94.36±3.24%,100%, respectively, have a evidently variance in6Gy group(υ=18, t=5.508>1.734,p<0.05). The peripheral blood nuclear cell’s and sperm’s comet rate is all100%,100%,have no-statistical significance in8Gy,10Gy group. Conclusions:The DNA of whole blood cell would damage by above2Gy X-ray. Nearly100%damaged would happen over6Gy X-ray, with repairing decrease d even disappeared. It’s suggested that6Gy is possibly the maximum dose the human whole blood cell can withstand. The sperm motility rate and sperm viability have no significantly change after different dose of radiation, also have the ability to combine with egg, but the sperm’s DNA was damaged badly. The sperm’s SCGE is more sensitive than peripheral blood nuclear cell’s SCGE in reflect the X-ray damage in a certain extent.
