Objective: To investigate the expressions of Nemo-like kinase (NLK) in BA(basilarartery) walls and cerebral cortex following subarachnoid hemorrhage (SAH) in SD rats andto study the relationship between NLK and cerebral vasospasm (CVS), and early braininjury (EBI).Methods:60health male SD rats were assigned randomly into following groups:Control group(only injected normal sodium through cisterna magna) and SAH group. SAHgroups then were assigned into5subgroups according to time points(1d,3d,5d,7d and14d, n=10). Once autologus arterial blood injection through cistema magna wasunderwent to establish SAH model in rats. By hemotoxylin and eosin(HE) staining, themorphologic changes of BA were examined and the expressions of NLK in BA walls andcerebral cortex following SAH at different time point were detected by usingimmunohistochemistry method,Western blot analysis and RT-PCR.Results: The pathological changes including thickness of arterial wall, arterialnarrowing, hypertrophy of smooth muscle cells and reductus of intima could be observedand such changes progressed with time and were most obvious on3rd day. Thecross-section area of BA on3th day after SAH was about43.5%of the control group’s, andthe wall thickness of BA was about1.7times of the control group’s. The manifestation on14th day was near to the control group. Western bolt analysis and RT-PCR show that theexpression of NLK in BA(basilar artery) walls and cerebral cortex decreased in SAH group,and reached to the bottom on3rd day and5th day, then increased. The level on14th daywas similar to the control group.Conclusions:1. The pathological changes including morphological changes,arterynarrowing, thickness of arterial wall shows that CVS occurs after SAH.2. In SAHgroup, the expression of NLK has negative correlation with CVS. It suggests that NLKmay participate the pathological course of vasospasm.3. In SAH group, the expression ofNLK in the cerebral cortex walls decreased first and then increased. It suggests that NLKmay participate the pathological course of early brain injury.
