In the meanwhile, toprovide evidence for the pathogenesis of RA, and the clues for the diagnose andtherapy.Methods1. Using the complete freund’s adjuvant and Col II inject DBA mouse, to set up mousemodel with RA.2. Collecting blood of patients with RA and separate single nucleus, and then select verall RNA and protein.3Using PCR detect RA mouse model and the spleen control group, and thedifference of gene expression of PCNP and TIPE2between normal persons andRA patients’ lymphocytes in blood.4. Using western blotting detect RA mouse model and the spleen control group, andthe difference of protein expression of PCNP and TIPE2between normal personsand RA patients’ lymphocytes in blood.5. Using real-time quantitative fluorescence PCR detect the difference of geneexpression of PCNP and TIPE2between RA patients and normal persons controlgroup’s lymphocytes in blood.Result1. Having been inducted DBA mouse model with rheumatoid arthritis successfully,and mission success rate is100%.2PCR has detected that the gene expression of PCNP and TIPE2in the spleen of RA mouse model and lymphocytes of blood of patients with RA is higher thancontrol group.3. Western blotting has detected that the protein expression of PCNP and TIPE2inthe spleen of RA mouse model and lymphocytes of blood of patients with RA ishigher than control group.4. Real-time quantitative fluorescence PCR has detected that the gene expression ofPCNP and TIPE2in RA patients is higher than control group, and the expressionquantity in active period is higher than stable period.Conclusion1. It is high expression that PCNP and TIPE2in the spleen of RA mouse model andblood of RA patients.2. The expression quantity of PCNP and TIPE2is related to severity of RA patients,and it may be an important index for diagnose, therapy and prognosis of RA.
