Hypoxia inducible factor(HIF)is a kind of transcription activator which helps most of organismsmaintain survival situation in anaerobic condition. In recent years, HIF has been a hot subject for manyexperts and scholars. Numerous studies proved that HIF plays an important role in tumour’s survival andevolution in anaerobic condition. The studies about the relationship between HIF and breast cancer mostlyconcentrate on these fields like survival of HIF-1α and breast cancer histiocyte, angiogenesis factor andglucose transporters, but lack of clinical pathology study in infiltration and metastases relationship ofHIF-1α and breast cancer.Objective The paper analyzes the correlation of metastases process between the expression of HIFand epithelial-mesenchymal transition,studies the effect of HIF on invasive ability and apoptosis of breastcancer cells, and assesses the relationship between hypoxia inducible factor and triple-negative breastcancer. At the same time, the paper will also provide full and accurate datum for clinical pathology study inHIF as candidating tumor maker.Methods Operative-move-out mammary gland sample were collected, fixied with formaldehydesolution, embeded with routine paraffin, sectioning, dewax and hydrate, classifying by diagnostic results;The protein expressions of HIF-1α of the breast carcinoma tissues, E-cadherin, Vimentin, CD34, ER, PRand Her-2were Observed by immunohistochemistry. Beside these, this paper also used TdT-mediateddUTP Nick-End Labeling(TUNEL)to test apoptosis of cells in breast carcinoma.Results HIF-1α positive expression rate related to clinical stages, i.e. the higher the rank of TNMwas, the higher the positive rate of cell was. The positive expression rates of HIF-1α patients who had metastatic lymph nodes of the breast cancer were obviously higher than the rates of those without lymphnode involvement; the positive expression rates of HIF-1α patients with negative expression of ER or PRwas higher than the rates of those who are with positive expression of ER or PR(P<0.05). The bigger thevolume of tumour was, the higher the rank of TMN and the value of MVD were. the value of MVD of thepatients with metastatic lymph nodes of the breast cancer were higher than those without lymph nodeinvolvement(P<0.05). As increasing of expression intensity of HIF-1α, the patients’MVD valueraised,too. Correlation analysis showed that they were positive correlation(r=0.862,P<0.01). Theexpression of E-cadherin in lymph node metastasis tissue was lower than that of the tissue had notmetastases(zc=4.36,P<0.01). The expression of Vimentin in lymph node metastasis tissue was clearlyhigher than that of the tissue had not metastases(zc=5.05,P<0.01). Consistency-check indicated thatHIF-1α and E-cadherin had good consistency,i.e. high correlativity(Kappa=0.793,P<0.01). HIF-1α andVimentin also had better relevance (Kappa=0.791, P<0.01). If HIF-1α is negative byimmunohistochemistry, the apoptosis’quantity of cancer cells of breast cancer patients was maximum. IfHIF-1α is positive by immunohistochemistry, apoptosis’quantity of cancer cells was less than the firstcase, but datum had no statistical difference(P>0.05). If HIF-1αis positive(++)by immunohistochemistry,the quantity of cancer cells’ apoptosis was less (P <0.05)than the quantity of the cells belonging to thosewhose HIF-1α was negative(-,+);But compared with patients whose HIF-1α are negative(-,+), theapoptosis’quantity of those whose HIF-1α was positive (+++) decreases evidently (P <0.01)。Thepositive expression rate of HIF-1α of patients in TNBC group was about61.40%; but in non-TNBC group,the expression of most patients’ HIF-1α was negative and the positive expression rate was only35.68%(zc=4.79,P<0.01).Conclusions 1. The positive expression of HIF-1α of the invasive ductal breast carcinoma correlates withpathological grading and lymph node metastasis.2. The positive expression of HIF-1α of the invasive ductal breast carcinoma and the marker ofepithelial-mesenchymal transition is correlates positively.3. Patients whose testing of HIF-1α of the invasive ductal breast carcinoma is positive, their quantity ofapoptosis of cancer cells decrease correspondingly.4. The positive expression rate of HIF-1α of TNBC patients is higher than non-TNBC patients.
