The Expression And Significance Of MiR-339-3p And MiR-339-5p In Vitro Gastric Carcinoma Cell Lines

Objective:To study the expression levels of miR-339-3p and miR-339-5p in the gastric carcinoma cell lines (SGC-7901,BGC-823and MKN-45) and gastric surface epithelium(GES-1);detect the relationship between miR-339-3p and miR-339-5p and the gastric carcinoma cell lines in vetrio experiment through the gain of function,and further significance is suggested.Methods:One Step PrimeScript miRNA cDNA Synthesis Kit and SYBR green I real time PCR were performed to access the expression of miR-339-3p and miR-339-5p in different cell lines.Results were counted by using2-ΔΔCt method to describe the differents between the gastric carcinoma cell lines (SGC-7901,MKN-45and BGC-823)and gastric surface epithelium (GES-1).The expression levels of miR-339-3p and miR-339-5p were verified by real time PCR experiment again after transfecting miR-339-3p mimics and miR-339-5p mimics. After that,we did the Annexin V-FITC apoptosis assay by flow cytometry and cell proliferation tests by CCK-8assay after72hours.Statistics was accomplished by Q test and paired t test.0.05levels was used for rejection of null hypothesis.Results:(1) Real time PCR results showed that the expression levels of miR-339-3p and miR-339-5p in gastric carcinoma cell lines was lower than gastric surface epithelium. Compared with GES-1, the expression level of miR-339-3p in SGC-7901,BGC-823, MKN-45was0.142,0.086and0.184respectively; also,the expression level of miR-339-5p in SGC-7901,BGC-823and MKN-45was0.115,0.091and0.261respectively.(2)miR-339-3p and miR-339-5p can be overexpressed by transfecting with miR-339-3p mimics or miR-339-5p mimics respectively.(3)After transfection with miR-339-3p mimics or miR-339-5p mimics,apoposis of MKN-45increased in contrast with negative control after72hours(P<0.05).(4)Compared to transfection negative control and the blank group,the ability of proliferation of MKN-45cell line decreased after transfecting miR-339-3p mimics and miR-339-5p mimics within72hours(P<0.01).Conclusion:(1) The expression levels of miR-339-3p and miR-339-5p significantly decreased in the gastric carcinoma cell lines in contrast with gastric surface epithelium. And the decline was not concerned in the cell differentiation.(2)miR-339-3p and miR-339-5p may be involved in the apoptosis and proferation of the gastric carcinoma.
