Objective:To observe the effect of combinating with using the large dose of γ-aminobutric acid(GABA), Sodiuln dimercaptopropanesulfona (NA-DMPS), vitB6and hyperbaric oxygen to acute the tetramine intoxication in rats, to explore the best clinical treatment of tetramine intoxication.Methods:Through the acute toxicity experiment, healthy female or male SD rats randomly divided into the control group (saline intraperitoneal injection of treatment), group A, group B, and group C, each group15rats. After60rats were given tetramine(0.35mg/kg) intragastric administration. Group A was given saline (7ml/kg) intraperitoneal injection and hyperbaric oxygen therapy;Group B was given5%GABA (350mg/kg), Na—DMPS (350mg/kg), vitB6(700mg/kg) intraperitoneal injection of treatment; Group C was given5%GABA (350mg), NA-DMPS(350mg), vitB6(700mg) intraperitoneal injection and hyperbaric oxygen therapy;The control group was given saline (0.7ml/kg) intraperitoneal injection.After administering, rats were recorded of breathing, observing the rat poisoning symptom, the incubation period of convulsions, the frequency of convulsion in1hour, the time of death, death numbers and mortality immediately. before putting to death or after administerring drugs (excluding the rats which died before doing hyperbaric oxygen therapy), the rats immediately collected arterial blood, which was detected alanine transarninase (ALT), aspartate aminotransferase (AST), creatine kinase (CK), creatine kinase-MB (CK-MB).RESULT:Compared with control group (saline water),1hour mortality of group A (saline+hyperbaric oxygen therapy) was significant difference (P<0.05);1,24hour mortality of group B (5%of GABA, NA-DMPS, vitB6) and group C (hyperbaric oxygen therapy+5%of GABA, NA-DMPS, vitB6) was significantly lower than control group and group A, there was statistically significant difference(P<0.05);In group B and group C, the latency for convulsion/muscular twitch was (9.7±3.6,10.3±3.8) min respectively,1hour interval time of twitch was (11.7±4.1,12.3±5.0) min respectively, the average time of death was (158.6±17.5,167.6±38.7) min respectively. Compared with control group and group A, above index of group B and group C was extended, there were significant difference(P<0.05). Compared with control group in latency for convulsion/muscular twitch,1hour interval time of twitch and average time of death, there were no significant difference in group A(P>0.05) Compared with control group, group B and group C in the serum levels of ALT, AST, CK and CK-MB significantly lowered (P<0.05), and those serum levels of enzymes were lower group C than those in group B(P<0.05). There were no significant difference in those serum enzymes between group A and control group(P>0.05).Conclusion:Treatment of hyperbaric oxygen alone has no certain effect. They were apparently effective in both the combination of large dose of GABA, NA-DMPS, vitB6and hyperbaric oxygen or the combination of large dose of GABA, NA-DMPS and vitB6, which treated acute tetramine intoxication in rats. The treatment using the combination with hyperbaric oxygen is superior to the treatment of the combination without hyperbaric oxygen.
