Objective:To observe the changes of nitric oxide, endothelial nitric oxide synthase (eNOS) in gastric antrum tissue and soluble endothelial cell protein C receptor (sEPCR) in the serum in the diabetic rats, and to investigate the relationship of the gastric motility change and gastric vasculopathy in the early stages of type2diabetes mellitus.Methods:A total of25male SPF SD rats were first divided randomly in two groups:control group (NC, n=10) and diabetic mellitus group(DM, n=15). The rats in the DM group were fed with fat-and caloric-rich diet and induced by intraperitoneal injection of streptozotocin (STZ). At12weeks, gastric emptying(GET1/2) and gastric emptying rate were measured in NC group and DM group by single photo emission computer tomography(SPECT). Fructosamine was employed to examine the serum level of GSP. Enzyme-linked immunosorbent assay was employed to examine the serum level of sEPCR. NO in gastric antrum tissue was detected by nitrate reductase, and eNOS observation in the gastric tissue was measured by immunohistochemical assay.Results:1. The gastric half-emptying of diabetic rats was30.82±12.56min, was significantly lower than in NC group(47.86±19.65min,P<0.01);The gastric emptying rate of DM was1.92±0.49%(VS NC group, P<0.01);2. NO in gastric tissue in DM group was30.32±2.04umol/l,was decreased than NC group(34.38±3.58umol/l, P<0.01); eNOS immunoreactive cells in gastric antrum tissue in DM rats were lower than NC group;3. The serum level of sEPCR was significantly higher in DM group(129.45±4.83ug/l) than in NC group (116.44±8.05ug/l, P<0.01);4. Spearman related analysis showed that there was positive correlation between gastirc half-emptying time and NO in gastric antrum tissue and eNOS expression level in stomach tissue;there was negative correlation with FBG, serum sEPCR. The FBG (X1), sEPCR (X2), NO (X3), eNOS (OD value, X4) as the independent variable of multiple stepwise regression analysis, the results showed that NO level in gastric tissue (Beta=0.358, P=0.02) and eNOS immunoreactive cells in gastric antrum tissue (Beta=0.589, P=0.001) were significantly associated with gastric emptying, R2=0.324,regression equation:Y=8.744+0.358X3+0.589X4Conclusion:Our results indicate that gastric emptying is accelerated in the early stage of DM. In this stage,vascular endothelium is impaired, and thus results in gastric motility disorders. If the protection to endothelial function in the point of view of the prevention and cure of diabetic gastric motility disorders, and it may be beneficial that the prevention and cure of diabetes complications.
