Objective:1. To investigate the changes in the numbers of endothelial progenitor cells (EPCs) in whole blood, heart,liver,lungs, brain, kidney in rats of different growth period.2.To discuss the changes in the function of endothelial progenitor cells (EPCs) in whole blood, heart,liver,lungs, brain, kidney in rats of different growth period.3. To explicit the changes in the number and function and degree of ageing of endothelial progenitor cells (EPCs) in whole blood, heart,liver,lungs, brain, kidney in rats of different growth period, In order to explore the EPCs may and age-related disease occurrence and development of relations and for the clinical application of EPCs looking for a more ideal seed cells lay the foundation.Methods:1. Wistar rats at0.5months,2months,6months,18months of ages were10in each group. Blood with EDTA.K.2was collected by abdominal aorta when rats were anesthetized by2%pentobarbital sodium.Meanwhile rats in each group to prepare heart, liver,lung,brain. kidney’s same place tissues and organs as single cell suspension, with CD34-FITC and CD133-PE double staining and detect the changes of the number of EPCs in blood and tissue suspension by flow cytometry. and take the same place of heart, liver.lung,brain and kidney in tissue microarray, detect the tissue staining of CD34and CD133EPCs by immunohistochemistry.2. The selection of Wistar6rats (4is female and2is male), do the latter training, after choose the child of three generations of rats to do the continue inbreeding, to do group raising when in the process of grows according to the experimental design needs to choose the health rats,according to the experiment scheme testing after arrived at the scheduled time.3. Use the enzymelinked immunosorbent assay (Elisa) to test the plasma VEGF and NOS level of each rat, detection heart.liver, lung, brain.kidney of the same parts organization and other heavy beta and a half the content of lactose enzymes of rat.Results: 1. The number of EPCs by flow cytometry of blood and heart, liver.lung.brain,kidney in2months and8months rats were significantly higher than0.5months and18months groups (P<0.05).but there are no significant in2months and8months groups(p>0.05). The same as0.5months and18months groups.2.The level of VEGF and NOS in plasma of older rats is decreased significantly, there are no significant in2months and8months groups(p>0.05).0.5months is highest level; The level of β-half of lactose glucoside enzymes is gradually increases in the heart, liver,lung,brain,kidney.Conclusion:1. The Flow cytometry detection technique:With the growth and development of the rats,The number of EPCs Of in the peripheral blood is regular change, suckling rat group and the elderly group less, but the young group and middle-aged group are more.2.The positive expression of EPCs by immunohistochemical staining of blood and heart, liver,lung,brain,kidney in2months and8months rats were significantly higher than0.5months and18months groups (p<0.05).but there are no significant in2months and8months groups(p>0.05). The same as0.5months and18months groups.2.Immunohistochemical detection displayed:With the growth and development of the rats, EPCs number of the major organs, including the heart, liver, lung, brain.kidney is a regular change, infants rat group and the elderly group is less,but the yong group and middle-age group is more.3. VEGF and NOS level of Plasma is gradually decreasing reaction the function of endothelial progenitor cells gradually decrease,(3-half of lactose glucoside enzymes level of same issue in the heart, liver, lung, brain,kidney is gradually increasing infect EPCs Constantly aging at the different development stages.4.0.5months with18months of group compared:the number of peripheral blood and main viscera endothelial progenitor cells although similar, but function of18months group is obviously a recession.
