Background:Vitexicarpin is one of the flavonoid components extracted from Vitex rotundifolia, many studies have reported that it can inhibit proliferation of kinds of tumor cells,however, the effect of vitexicarpin on pituitary adenoma cell has not been reported. Objectives:To investigate the anti-proliferration effect on pituitary GH3cells and explore the molecular mechanism. Methods:We evaluate the proliferation of GH3cells treated by different solubility gradient of vitexicarpin by drawing the cell viability curve.We used CCK-8method to evaluate the time-and dose-dependent effects. The effect of vitexicarpin on GH3cell apoptotic and the change of apoptotic protein Bcl-2,Bax,PARP,Caspase3were evaluated by using flow cytometer and western blot respectively.Therefore,the molecular mechanism of vitexicarpin on GH3cell was clarified. Results:Vitexicarpin can inhibit the growth and proliferation of GH3cells and this inhibitory is a time-and dose-dependent effect. the results of flow cytometry showed that vitexicarpin can promote GH3cell apoptosis. Treated by vitexicarpin, the Bax protein level was significantly increased;Bcl-2was lower; caspase-3was activeded and PARP was degradated.Conclusions:We concluded that vitexicarpin can inhibit proliferation of GH3cell and mitochondria dependent apoptosis may partly be the mechanism.
