Liver is the key organ for metabolism,because it is responsible for most of theanabolism, catabolism and transforlation in vivo,so it’s essential to investigate metabolicmechanisms of liver damage.Metabonomics study focus on the metabolites and intermediatemetabolites,makes a complete metabolite profiling analysis, Adequately reflect the biologicalsystem’s response to the gene and the external environment changes,further clarify metabolicpathways.Nowadays,metabonomic studies on the hepatic injury have achieved initialprogress,but not yet in the pediatric field.So we established an metabonomic researchplatform to look for biomarkers of infectious hepatic injury on children.Our study has built a metabolomics research platform applying HPLC-MS/MS,different analysis was utilized to compare the serum metabolic profiles of patientsand controls respectively,and then the potiential biomarker were further be found.30patients and30controls were enrolled in this study,the blood sample of the subjectswas taken in the morning,and liver function test was obtained from every patient.Pretreatment of serum sample including protein sedimentation was conducted beforechromatography. After imported into Markerview software,the raw file was manipulatedthrough peaking finding,fitering and peak alignment to obtain peak information.The resultwas exported to a two-dimensional table,which was further processed according to “80%rule”and total peak area was normalized to a fixed level,and then the data was disposed bymethod of PCA combined with t-test,cluster analysis,and SAM. Biological markers werescreened by data mining.The serum metabolic profiles between patients and controls have evident varience,23biomarkers were finally identified by PCA combined with t-test,45metabolites in differencewere displayed clearly by the drawing of cluster analysis,and9biomarkers were found bySAM.
