This thesis is divided into four parts:Chapter1Literature ReviewHypoglaucous Collett Yam Rhizome is the dried rhizome of the Dioscorea collettii Hook. f. var. hypoglauca(Pslibin)Pei et Ting. It’s bitter, flat, and belong to kidney, stomach meridian, expelling wind Tongbi, dampness go cloud effect for gonorrhea gonorrhea, waist and bi disease, damp, sore, vaginal discharge excessive Fengshibitong bad for the jointscertificate of treatment. The major chemical constituents, pharmacological effects and clinical applications of the Dioscorea collettii plants were reviewed. It contains draw-steroidal (including Lo sterane class contains Dioscorea collettii furostanol alkanes pregnant sterane, cholestane glycosides, and phytosterols), the diaryl heptane class, lignans, organic acids and the lipids and other chemical ingredients. With blood pressure, uric acid, anti-inflammatory and analgesic, antibacterial, improve immunity, anti-tumor, kill molluscs and other pharmacological effects. In clinical, it has been used for the treatment of gonorrhea gonorrhea, excessive vaginal discharge, sore and hot and humid, waist and knee pain and rheumatism Bi stubborn and difficult urination and other symptoms.Chapter2Study on the chemical composition of the Hypoglaucous Collett Yam Rhizome19compounds were obtained from the Hypoglaucous Collett Yam Rhizome by means of a variety of chromatographic techniques,16compounds were identified by variety spectrum. Including8steroidal saponins and glycosides, named diosgenin, dioscin, gracillin, dioscin secondary saponins A, protodioscin, methyl protodioscin, protogracillin, methyl protogracillin;2Diaryl heptane class, named1,7-bis(4-hydroxyp henyl)-1,4,6-heptatrien-3-one and1,7-bis(4-hydroxyp henyl)-4,6-heptabien-3-one;1Alkaloids, named trichosanatine;1Philippines and pyran derivatives, named Dioscorone A;1Coumarins, named,(3S)-6,8-two hydroxy-3-phenyl-3,4-dihydro isocoumarin;2Sterol glycosides,named, β-sitosterol, Daucosterol;1Fatty acids,named, Palmitic acid.1,7-bis (4-hydroxyp henyl)-1,4,6-heptatrien-3-one and1,7-bis(4-hydroxyp henyl)-4,6-heptabien-3-one, trichosanatine, Dioscorone A,(3S)-6,8-two hydroxy-3-phenyl-3,4-dihydro isocoumarin and Daucosterol were obtained from the Hypoglaucous Collett Yam Rhizome for the first time.Chapter3Method for the determination of steroidal index components of the Hypoglaucous Collett Yam RhizomeHPLC methods were established for the determination of protodioscin, diosgenin. The regression equations of these four compounds were as follows Y=111214X-23782(r=0.9991), Y=143558X+1721.9(r=0.9996), respectively. The linear ranges of the protodioscin, diosgenin1.728~8.640μg,1.025~8.200μg respectively. The average recoveries for protodioscin, diosgenin were101.98%,101.60%, RSD were1.53%,2.41%(n=6) respectively. The results of the contents of these two compounds in10batches of raw drug showed the methods were reliable, accurate and able as the Hypoglaucous Collett Yam Rhizome herbs of protodioscin, diosgenin constituents content determination methods.Perchloric acid colorimetric were established for the determination of the total saponin content of the Hypoglaucous Collett Yam Rhizome herbs diosgenin as reference, the measured absorbance at408nm at the color diosgenin standard curve was Y=0.0886X-0.0075, r=0.9995(n=6), showed that the diosgenin absorbance linearity in the range of1.55~12.42μg/mL is good.2.76%of total saponins was determined that the Hypoglaucous Collett Yam Rhizome herbs (Nanning, Guangxi), and RSD was1.16%, the recovery was100.69%, RSD2.75%(n=6).4origin of the Hypoglaucous Collett Yam Rhizome herbs total saponins were measured, the results show that the method is simple, accurate, measured as the Hypoglaucous Collett Yam Rhizome herbs total saponins method.Chapter4Summary and DiscussionInnovation:1.5compounds were obtain from the Hypoglaucous Collett Yam Rhizome for the first time, they are1,7-bis (4-hydroxyp henyl)-1,4,6-heptatrien-3-one and1,7-bis (4-hydroxyp henyl)-4,6-heptabien-3-one, Dioscorone A,(3S)-6,8-two hydroxy-3-pheny1-3,4-dihydro isocoumarin and Daucosterol;1compound was obtained from the Dioscorea Linn for the first time, it is Dioscorea Linn.2. The HPLC method for simultaneous determination of Protodioscin, diosgenin in the Hypoglaucous Collett Yam Rhizome was established for the first time. Provide a prove for comprehensive and accurate evaluation of the Hypoglaucous Collett Yam Rhizome quality.3. The method for the determination of the Hypoglaucous Collett Yam Rhizome total saponins was established for the first time.Measured the Hypoglaucous Collett Yam Rhizome total saponins by perchloric acid colorimetric,as the basis for the quality control of the Hypoglaucous Collett Yam Rhizome.
