ObjectiveTo investigate whether octreotide, as somatostatin analogue, can enhance the sensitivity ofthe human lung adenocarcinoma A549cells to chemotherapeutic drugs.MethodsThe absorbance value was tested by colorimetry through MTT method to evaluate the effectof octreotide, cisplatin,5-fluorouracil or the three drugs combined respectively after48hours.Each drug concentration in six holes and it repeated three times. The effects of combinationtherapy is analysed with Isobologram.ResultsIt was proved that Octreotide could inhibit the proliferation of A549cells in adose-dependent manner at the concentration rage of1.3mg/L～166.7mg/L. The inhibition washigher when octreotide was combined with cisplatin and5-fluorouracil than it alone. It hasstatistically significant difference（P<0.05）.The effect plots of IC50was located in the synergyareas of isobologram.ConclusionIt can be concluded that Octreotide could inhibit the proliferation of A549cells in vitro.This inhibition enhances when octreotide is combined with cisplatin and5-fluorouracil..Octreotide can enhance the susceptibility of A549cells to cisplatin and5-Fluorouracil. Octreotide in theS and G2phase cells increased, the DDP cells in G1phase increased5-Fu, cisplatin combinedwith5-Fu and three-drug combination role of the cells arrested in S phase. Late apoptosis ofcisplatin combined with5-Fu and three-drug combination effects on A549cells was significantlyhigher than the effect of the drug alone, there are significant differences (P <0.05); three-drugcombination compared with cisplatin combined with5-Fu. significant difference (P<0.05).Western blot results show the three drugs in A549cells after p53protein expression isincreased in the combined effects of three drugs the highest expression of p53protein incombination with cisplatin5-Fu there was significant difference (P <0.05)..ConclusionThe flow cytometry results showed that the three drugs have an effect on A549cell cycleand apoptosis, the combined effects of three drugs, the most obvious effect of p53protein isinvolved in the drug combined effects of the process.
