ObjectiveTo observe the effect of TGF-βand ultra microstructure changes in pediatric unclosedprocessus vaginalis and peritoneum and to increase the cognition of pediatric indirect hernia,hydrocele pathogenesis and clinical treatment.MethodsTake 30 cases of operational excisional hernia sac and Hydrocele sac and 10 cases ofperitoneum of non-phlogistic patients to observe the changes in histopathology by using HEstaining. TGF-βamong them was detected by immunohistochemistry staining. Finally,weobserve their microstructure with.electron microscope. All the data of experiment wereanalyzed by statistics software SPSS17.0.Results(1) Morphological changes:The surface of natural peritoneum in collator was constructed withmesothelium,and there were mainly fibrillar connective tissue and fat and blood vessel and soon,and smooth muscle. We can observe 22%of them with phlogistic cells infiltration ,12%ofthem with fibrous tissue proliferation clearly,and glassydegeneration in the collator of indirecthernia and in the hydrocele of tunica vaginalis.(2) Expression of TGF- proteins by immunohistochemical staiming: TGF- were diffusedmoderate dyeing in indirect inguinal hernia,hydrocele and peritonealgroup(50.0%,50.0%,46.6%);while TGF- was not statistically significant in every group(P>0.5).(3)Ultra microstructure:The surface of natural peritoneum was constructed withmesothelium.The cell nucleus were flat and anomalistic.There were a lot of microvilli in theceliac surface,pinosome in mesothelium. and mainly fibrillar connective tissue and fat andblood vessel and so on under mesothelium.Compared with peritoneal, based on the observationof specimens under electron microscope its surface microvilli disappeared in 1 case;mesothelial cell shed, even disappear,only the basement membrane or fibrous connective tissuecan be seen in 3 casea; there were a lot of smooth muscle in 1case; its surface microvillidisappeared and mesothelial cell and fibrous connective tissue can be seen in 2 cases of thehydrocele of tunica vaginalis.ConclusionThe hernialsac of child indirect inguilal hernia and hydrocele of tunica vaginalis were bothconstructed by peritoneum.TGF-βhas no direct relation in the occurrence and developingprocess of pediatric indirect inguinal hernia and hydrocele.The hardcore of non-operationtreatment was considered that the formation of the aseptic inflammation can lead celluloseadjoin,and making processus vaginalis closed finally. In this study,we can observe many phlogistic cells infiltration ,fibrous tissue proliferation clearly. Compared with peritoneal, basedon the observation of specimens under electron microscope its surface micro villi disappeared;mesothelial cell shed, even disappear; In some cases only the basement membrane or fibrousconnective tissue can be seen.In addition,in some cases,there were a lot of smoothmuscle.Varying degree changes can be seen in hydrocele group. It was brought by the frictionof the cytoplasm. Peritoneum was easily led cellulose adjoin during its damage repair.Thischange hinted that it may relate with processus vaginalis closed. In addition,in some cases,smooth muscle can be seen.This hinted that it may relate with heinia develoption.
