Objective Observe the gross and histological changes in rats with deep partial thickness burnafter using collagenase application ointment locally. Detect EGF, bFGF expression in woundrepair processing.Then preliminary discuss the effect of collagenase ointment on the woundrepair processing and its mechanism.Methods20male SD rats were numbered, each rat were made two wounds in the back, Selectedone randomly as experiment group, the other as control group. The experiment group was treatedwith collagenase ointment, the control group with vaseline gauze.1/2d, Until wound healing.Observe the wound exudates, crusta formation and separating degree every day, Recording thehealing time, Measuring the healing area periodically to calculated the rate of healing. Catch onegroup with5rats one time in7,10,14,21d.tissue of each group was collected from the edge ofwound. The tissue was made into pathological and EGF, bFGF immunohistochemicalsections,The number of positive cells was counted in high magnificationThe experimental dataexpressed with x±s, using SPSS17.0statistical software to processing, Comparision betweengroups using the paired t test.Results10d The experimental group complete debridement cases was5, control group1cases.14d for experimental group13cases, control group6cases.17d for experimental group20cases, control group11cases.The wound healing time of experiment group was20.75±1.71dsignificantly shorter than that in the control group25.25+1.71d (t=6.971, P=0.006).7,10,14,21d healing rates were37.60+7.42,54.52+7.20,77.72+8.50,98.04+3.92, higher thanthat of control group19.06+4.26,35.96+5.88,49.48+6.04,81.80+6.04(t=8.200,9.116,10.102,5.294; P <0.05). Pathological examination showed that the experimental group is betterthan the experimental group in the quality of wound healing. Immunohistochemistry revealedthat the positive expression of EGF in the experimental group were higher than that of controlgroup (t=2.671,3.127; P=0.022,0.010) in7d14d, and significantly higher than the controlgroup (P=6.585, P <0.001) in10d.The expression between experimental group and controlgroup had no difference (t=0.309, P=0.763). The positive expression of bFGF in theexperimental group were higher than that of control group (t=2.370,2.918; P=0.037,0.014)in7d14d, and significantly higher than the control group (P=9.106, P <0.O01)in10d, Theexpression between experimental group and control group had no difference (t=0.132, P=0.898)Conclusion collagenase ointment promote wound repair in rats with partial-thickness burn, itsmechanism was relatedt to that collagenase can accelerated the separation of necrotic tissue,reduced local inflammatory response, increased the expression of in wound.
