ObjectiveTo explore the expression of TGF-betal in Ocular Surface with dry eye patients and study the correlation with the dry eye’s classification, synptoms and signs.MethodsIn this case-control study,30dry eye patients(30eyes) and15control subjects(15eyes) underwent full ophthalmologic examination (including OSDI questionnaire, tear break-up time, Schirmer’s test, corneal fluorescence staining and conjunctival lissamine green staining),and mild dry eye group(18patients18eyes) or moderate/severe dry eye group(12patients12eyes) were divided by OSDI questionnaire. Impression cytology was used to collect conjunctiva epithelial cells of dry eye patients and age-matched controls, then these samples were stained by IHC reaction and were detected the expression of TGF-betal by real-time PCR.Results1. The average OSDI questionnaire scores of mild group,moderate/severe group and control were22.61±2.20,44.67±7.58,9.00±3.34.There were significant difference in different degree of dry patients and control subjects(P<0.05).The average Sit of mild group, moderate/severe group and control were7.33±1.97,3.58±1.62,10.40±2.69. There were significant difference in different degree of dry patients and control subjects(P<0.05). The average TBUT of mild group, moderate/severe group and control were7.50±2.18,3.58±1.16,11.20±2.21. There were significant difference in different degree of dry patients and control subjects(P<0.05). The average corneal fluorescence staining of mild group, moderate/severe group and control were2.67±1.68,5.83±1.99,0.60±0.51. There were significant difference in different degree of dry patients and control subjects(P<0.05). The average conjunctival lissamine green staining of mild group, moderate/severe group and control were4.44±1.98,6.25±1.96,1.93±1.58. There were significant difference in different degree of dry patients and control subjects(P<0.05).2. The average expression of TGF-betal of mild group, moderate/severe group and control were1.40±0.20,2.07±0.42,0.73±0.42. There were significant difference in different degree of dry patients and control subjects(P<0.05). 3. The expression of TGF-betal had a positive correlation with OSDI questionnaire(r=0.814,P<0.05), corneal fluorescence staining(r=0.748,P<0.05), conjunctival lissamine green staining(r=0.590,P<0.05),and had a negative correlation with TBUT (r=-0.693,P<0.05) and Schirmer’s test(r=-0.645,P<0.05).ConclusionIt’s a effective way to research pathogenesy of ocular surface disease through impression cytology combination with IHC staining or real-time PCR. The IHC staining show that there are more TGF-β1in conjunctival epithelial cells with the dry eye patients than control subjects.The result of real-time PCR detected that there are obviously more TGF-β1in conjunctival epithelial cells with the dry eye patients than control subjects. The expression of TGF-betal has a positive correlation with OSDI questionnaire, corneal fluorescence staining and conjunctival lissamine green staining, but has a negative correlation with TBUT and Schirmer’s test. The expression of TGF-betal in different degree dry eyes appears to reflect the disease’s progression, then provode a reference for the diagnosis and the classification.
