Objective:This study used proteomic fingerprint technology, combining the magnetic beads with matrix-assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF-MS),to screen for potential protein biomarkers of the acute myeloid leukemias (AML) and AML-M3to establish and verify a diagnostic model, detect minimal residual disease (MRD) of AML-M3associated proteins.Methods:Serum proteins from110primary patients with AML(45cases of AML except M3,36cases of AML-M3,29cases of M3during complete remission) and32healthy controls were profiled and compared.The spectra were analyzed statistically using flexAnalysisTM and ClinProtTM bioinformatics software.Results:We build5models. Comparison of the protein profiles between5models resulted in44,45,34,4and7most significant peaks between primary AML/healthy control, primary AML except M3/healthy control, primary M3/healthy control, primary M3/primary AML except M3, primary M3/M3CR, respectively, were selected out using the ClinProTool software package and used to train a QuickClassifier algorithm(QC). Statistical analysis revealed that three peptides with m/z4642,7764and9289Da(P<0.000001) were considered as the protein biomarker of primary AML and M3MRD. Two protein m/z peaks at2723Da、6189Da(P<0.0001) that were highly expressed in primary M3, rather than other subtypes, were used to discriminate primary M3from AML. A diagnostic pattern for M3CR using m/z4642、9289Da(P<0.000001) was established. Blind validation yielded correct classification of6out of8cases.Conclusions:The MALDI-TOF MS analysis of AML patients serum protein indicated that the pattern of combined markers potentially provides a powerful and reliable diagnostic method for primary AML and primary AML-M3, and a promising dynamic method for M3MRD detection; the two peptides with m/z4642、9289Da may be better biomarkers for M3MRD.
