Objective:1:By Intranasal administration of low dosage recombinant human erythropoietin (rhEPO)in acute cerebral ischemia,to show that the rhEPO intranasal administration played a cerebral protective effect on cerebral ischemia in rats, intranasal application of rhEPO is an effective administrationmanner.2:Observation of rhEPO on the expression of acute cerebral ischemia in the rat brain matrix metalloproteinase-9(MMP-9), caveolin-1(cavolin-1Cav-1), explore the neuroprotective mechanisms of rhEPO, to provide the experimental basis for clinical application of rhEPO.Methods:ninety healthy male SD rats were randomized into sham-operated group,Ischemia group and rhEPO treatment group. Suture method was used to establish permanent middl e cerebral artery occlusion (pMCAO) model. rhEPO treatment group was given48UrhE PO via intranasal administration after ischemia for1hours, ischemia group andsham-oper ated group were given the same volume salinen at the time.All rats were decapitated afte r ischemia for24hours, and water contents in the brain tissues were calculated by dry-wet weight method, the permeability of blood-brain barrier were measured by method of Evans blue flurometry, the infarction volume was measured by TTC staining (2,3,5-triphe nyl tetrazolium chloride), and the expression of matrix metalloproteinase-9(MMP-9) and cavolin-1(CAV-1) were detected by immunohistochemistry.Results:1:Cerebral ischemia and hypoxia, the brain tissue water content、the EB content and the volume of infarction increased, compared with the ischemia group, rhEPO treatment group showed that brain water content, EB content and volume of infarction were significantly reduced, the difference was statistically significant.2:Sham operation group see a slight expression of MMP-9and CAV-1, compared with the sham-operated group, the expression of MMP-9and CAV-1of ischemia group in immunohistochemistry method showed increasing, compared with the ischemia group, rhEPO treatment group via Intranasal administration showed lowered expressions of MMP-9but elevated expression of Cav-1.
