ObjectiveTo study the effect of extracellular ATP on the FoxO3a pathway of denervated gastrocnemiusmuscle cell.Methods54 female adult Wistar rat were randomly divided into three groups.Model of denervatedgastrocnemius muscle was set up after sciatic nerves of inferior border piriformis were transectedat the right side.For ATP treatment group, 0. 1 mg /dayATP was injected into the gastrocnemiusmuscle at the right side and the same amount of normal saline at the left side for 4 weekcontinuously.Both side of gastrocnemius muscle was injected the same amount of normal salineas control group.The normal control group(0 day) don’t do anything .At 0,2,7,14,28 daypostoperatively, Rat were killed and both side of gastrocnemius muscle were removed .Wetmuscle were wejghted and wet muscle ratio(right/left) was calculated.The expresstion ofp-FoxO3aS253, FoxO3a, , MAFbX protein and mRNA were detected using Western Blotting andRT-PCR respectively.ResultCompared with control group, the wet muscle ratio of ATP treatment group increased,thedifference was Statistically significant in different period of times(p<0.01).The expression ofFoxO3a mRNA and protein was lower than the control group, and the difference was notatistically significant (p>0.05) .The expresstion of MAFbX mRNA and proteion was the same asthat FoxO3a,but the difference was tatistically significant(p<0.05) .The p-FoxO3a proteinexpresstion remarkly increased in the ATP treatment group,the difference was Statisticallysignificant in different period of times when compared with control group(p<0.05).ConclusionExtracellular ATP maybe make the FoxO3a phosphorylated and then inhibited the expresstionof MAFbX proteion to delay the denervated muscular atrophy.
