| [Objective]In order to offer the inspiration on deeply bone marrow CD34+cells replication research invitro,we investigated the proliferative effection of bone marrow CD34+cells by Bone marrowMesenchymal Stem Cells (BMMSCs).[Methods]êVarioMACS was used for sorting of bone marrow CD34+cells, and then the bone marrowCD34+cell purity was tested by FCM. áIsolating Marrow Mononuclear Cells from abortion fetalbone explored by density gradient centrifugation, then the BMMSCs further extracted from themarrow mononuclear cells, and cells cultivation processed with combination of the explant-attachedmethod, FCM test was demanded for the identification of marker on the cultivated cells surface. The bone marrow CD34+cells were cultivated in Transwell chamber, then the active cells numberwas counted facilitated by atomic calculating instrument. MTT colorimetric method was adoptedfor the proliferation activity detection of bone marrow CD34+cells.[Results]êFCM assay showed high purity of the sorted bone marrow CD34+cells, the proportion ashigh as90%and even more. áFlow cytometry assay illustrated all the cultivated BMMSCs highlyexpressed with the cell surface marker CD44,CD29, but negative expressed with HLA-DR, CD45and CD34.The results reveal the purity of cultivated BMMSCs was in a very high degree. Theprogression of inverted microscope observation and atomic cell calculating followed with thebone marrow CD34+cells cultivation in Transwell cell, showed the amount of the active marrowCD34+cells in test group is higher than control group with a significant difference(P<0.05), andMTT colorimetric testified the bone marrow CD34+cells proliferation activity is higher in testgroup compared with normal group, which showed a significant statistical implication(P<0.05).[Conclusion]BMMSCs play important role in promoting activation of bone marrow CD34+cells, whichmaybe closely related with secretion of cell factors. |
