| Objective To study the expression of microRNA-223（miR-223）in diffuse large Bcell lymphoma (DLBCL), and its relationship to subgroups and prognosis.Methods 45 cases of DLBCL with follow-up data were studied by usingimmunohistochemical EnVision method for CD3,CD10,CD20, bcl-6 and MUM1. TheDLBCL were classified into germinal center B cell- like (GCB) and non-germinal centerB cell-1ike (non-GCB) subtypes according to Hans’algorithm. Agilent HumanmiRNA Microarray 16.0 was used to select the Micro-RNAs on 24 casesparaffin-embedded tissue of DLBCL with the follow-up data. The expression levels ofmiR-223 were examined by TaqMan real-time polymerase chain reaction ( RT-PCR).14 cases of reactive lymph node were selected as control.Results n 45 cases of DLBCL, 35.6% of cases were GCB and 64.4% werenon-GCB subtypes. The expression of miR-223 was 19.8 in GCB subtypes, while 15.8in non-GCB subgroups. There is no significant difference between GCB and non-GCBsubgroups (P=0.236). The expression of miR-223 was up-regulated in DLBCL, and was17.2 times of the reactivated lymph node.There is a significant difference between twosubgroups (P=0.014). The high expression groups of miR-223 was significantlycorrelated with longer over survival (OS P=0.011). Multivariate Cox proportionalhazard regression analysis revealed that they were independent poor prognosis factors,including the low expression of miR-223 (RR=5.445, 95%CI, 1.555-19.068, P=0.008) ,abnormal level of LDH (RR=3.974, 95%CI, 1.191-13.266, P=0.025), and IPI≥3(RR=4.044, 95%CI, 1.233-13.264, P=0.021) in 45 cases of DLBCL.Conclusions The expression of miR-223 has no relationship withimmunophenotypes, while the high expression group of miR-223 has longer OS thanthat in low expression group. These results suggested that miRNA-223 might be one ofthe new prognostic marker. |
