BackgroundPelvic inflammatory disease(PID) is a common disease among women, it refers to the upper female genital tract in a group of infectious diseases, which including uterine myositis, endometritis, fallopian tube inflammation, ovarian inflammation, pelvic connective tissue inflammation and pelvic peritonitis. If pelvic inflammation can not be timely and correct treatment, it may brings to chronic pelvic inflammatory disease, and may spread to the uterus, fallopian and other organs, even leading to female infertility, tubal pregnancy or other complications. PID seriously endangering the health of women.Pelvic organ prolapse (POP) is a common clinical disease in old women, means for a diseaase caused by the disfunction of pelvic floor muscle, fascia and ligament of uterus.POP often associated with stress urinary incontinence (SUI), They were referred to as the female pelvic floor dysfunction (FPFD). PFD can seriously impact on women’s health and quality of life. Most studies suggest that, the decrease of the total collagen content, collagen fibers and the decreased numbers of the ligament of pelvic connective tissue resulted in, what’s more, the fascia and other support structure relaxation, which leads to the occurrence of PFD. But at present the occurrence and development mechanism of FPFD is not very clea. It is very important for us to study its incidence mechanism of PFD diagnosis.Inflammatory mediators are a group of biologically active substances which can mediated inflammatory response. Studies shows that, inflammatory cytokines originate from different tissues and cells will appear in the repair process of a damaged ligament. Prostaglandins E2(PGE2) and interleukin1-β(IL1-β) are important inflammatory mediators, they play an extremely important role in mediating inflammatory reactions in the occurring, the development and the outcome of inflammatory.Previous studies suggest, prostaglandin E2and interleukin1-β do not only mediates inflammation in the occurrence and development of a variety of cells, cell proliferation and biological function of regulation.Through our study of prostaglandin E2and interleukin1-β in the separate and combined application on in vitro uterine ligament fibroblast proliferation activity in vitro, we’d like to understand the relationship of pelvic inflammation and the repair of impaired uterine ligament repair process.ObjectiveTo investigate the effects of inflammatory mediator prostaglandin E2(PGE2) and interleukin1-p(IL1-β) on cell proliferation in rabbit uterine ligament fibroblasts.MethodsThe third passage of rabbit uterine ligament fibroblasts were treated with PGE2(25、50、100、500、1000ng/ml) and IL1-β(10、20、50、100、200ng/ml) and PGE2+IL1-β respectively. Cell proliferation was measured by MTT method.Results The proliferative effects in100ng/ml PGE2and20ng/ml IL1-β inflammatory mediator group was statistically higher than that of control group and other dosage inflammatory mediator groups (P<0.05), and the combined group of PGE2(100ng/ml)+IL1-β(20ng/ml)showed an even higher proliferative effect than other groups (P<0.05).ConclusionThe single and combined use of inflammatory mediator PGE2and IL1-β in proper concentration significantly enhances cell proliferation of ligament fibroblasts,While the excessive use of inflammatory mediator restrains cell proliferation.It shows that the inflammatory factor PGE2and IL1-β indirectly regulate the cell proliferation of uterine ligament fibroblasts.
