| Objective:To investigate the effect of tripterygium wilfordill (TW) on the expression of transforming growth factor-β1(TGF-β1) and platelet derived growth factor-BB (PDGF-BB) in cultured rat glomerular mesangial cells stimulated by high glucose. Methods:To culture the rat glomerular mesangial cells in vitro, which is selected as research subject, as it is divided into the following groups after the subculture:normal glucose(5.6mmol/L),high glucose(30mmol/L), high glucose plus tripterygium wilfordill at5mg/L,10mg/L,20mg/L) for periods of12,24,48hours, respectively. The expression levels of TGF-B1and PDGF-BB mRNA in cells of each group was measured by RT-PCR, and the protein expression of TGF-B1and PDGF-BB in cells of each group was measured by immunocytochemistry. Results:1.The rat glomerular mesangial cells of normal groups all have the expression of TGF-β1and PDGF-BB;2.Compared with the normal glucose groups, the high glucose group cells has an increased expression of TGF-β1and PDGF-BB mRNA and protein (P<0.05);3. Compared with the high glucose group,the group of tripterygium wilfordill polyglucoside has a decreased expression of TGF-β1and PDGF-BB mRNA and protein (P<0.05)Conclusion:1.The rat glomerular mesangial cells have the expression of TGF-β1and PDGF-BB;2. The expression of TGF-β1and PDGF-BB mRNA and protein is increased in rat glomerular mesangial cells stimulated by high glucose;3.Tripterygium wilfordill can reduce the expression of TGF-β1and PDGF-BB in cultured rat glomerular mesangial cells stimulated by high glucose. |
