Objective:To observe the effects of shanmai capsules on the blood pressure in renovascular hypertensive rats by "two-kidney one-clip"method and on vascular endothelial function, study the antihypertensive effect of shanmai capsules, and discuss the mechanism of shanmai capsules promote antihypertension from regression of atherosclerosis.Methods:(1) Established renovascular hypertensive model in rats by "two-kidney one-clip"method.(2) To observe the effects of shanmai capsules on the blood pressure in renovascular hypertensive rats. Measured the systolic blood pressure (SBP) of caudal artery in rats by noninvasive blood pressure measurement system at before, after modeling and intragastric administration four weeks. Collect plasma and heart specimens of rats in the fourth weekend, measured the level of nitric oxide (NO), angiotensin Ⅱ(Ang Ⅱ) and endothelin (ET), and weighting the heart weight(HW)and left ventricular weight(LVW), measured remodeling of myocardial fiber diameter (MD) and renal arterial lumen diameter(RAD), luminal cross-sectional area(Lcsa) and media thickness(Mt) by pathologically method.(3) To observe the effects of shanmai capsules containing serum in different concentrations on routine culture Human umbilical vein endothelial cells (HUVECs) after24h and VCAM-1, ICAM-1were induced in HUVECs by tumor necrosis factor-α (TNF-α). Measured the level of NO, eNOS, VCAM-1and ICAM-1in cell culture fluid by Enzyme-linked immunosorbent assay (ELISA) method.Results:(1) The blood pressure of rats after renal artery narrow operation was significantly higher than blood pressure before operation.(2) Compared with model control group, the hypotensive effect of shanmai capsules on renovascular hypertensive rats was reversible and concentration dependent, not only reduced the level of Ang Ⅱ and ET in blood plasma and HW, LVW, MD and Mt, but also increased NO, RAD and Lcsa at the same time, differences have statistical significance.(3) Shanmai capsules containing serum not only increased amounts of NO and activities of eNOS in HUVECs obviously, but also reduced amounts of VCAM-1and ICAM-1at the same time, there were significant difference as compared with the model control group.Conclusion:(1) Renovascular hypertensive model in rats could imitate hypertensive disease of human. The method is a simple technique, with strong feasibility, high successful rate, and suit needs of empirical study of hypertension.(2) Shanmai capsules could reduce the blood pressure of renovascular hypertensive rats, inhibited the formation of myocardial hypertrophy and renal arterioles remoderling.(3) Shanmai capsules could increase eNOS activities in HUVECs, rusult in produced more NO, and also reduced the level of inflammatory mediator about VCAM-1and ICAM-1, protecte vascular endothelial function so that achieve regression of atherosclerosis. |
