Objective:To explore the therapeutical effect and its mechanism of Lycium barbarum polysaccharides(LBP) in oligozoospermia mice model,thus to provide the basis for its application in clinical therapy of oligozoospermia.Methods:SPF male Kunming mice were randomly divided into the normal group, the model group, positive control group, and the treatment group.Oligozoospermia model mouse was produced by intraperitoneal injection of cyclophosphamide,while the high,medium and low dose treatment group was fed high, middle and low dose of LBP respectively and the positive control group was fed clomifene citrate Aqueous solution. All mice were executed on the35th day. Such values were observed as the content of testosterone (T), luteinizing hormone (LH) and follicle-stimulating hormone (FSH) by electrochemiluminescence;the testis and epididymal index; the density of sperm;pathological changes of testis by HE staining light microscopy and immunohistochemical detection of each group in mouse spermatogenic cell expression of bcl-w and bax expression; And use the diagraph-analysis system to analyze them.Results:Compared with the normal group, the density of T in model group decreased (P<0.01), LH, FSH increased (P<0.01); the testis and epididymal index decreased (P<0.01); sperm density decreased (P<0.01); the epithelial layer of contorted seminiferous tubules and the number of spermatogenic cells decreased (P<0.01); Bax and Bcl-w expression of IOD value ratio in spermatogenic cells increased (P<0.01). Compared with the model group, the density of T in treatment group increased,LH, FSH decreased (P<0.01); the testis and epididymal index increased (P<0.01); sperm density increased (P<0.01); the epithelial layer of contorted seminiferous tubules and the number of spermatogenic cells increased (P<0.01); Bax and Bcl-w expression of IOD value ratio in spermatogenic cells decreased (P<0.01). Compared with the positive control group,the testis and epididymal index,the density of T,LH, FSH in treatment group were no significant difference(P>0.05);sperm density increased (P<0.01); the epithelial layer of contorted seminiferous tubules and the number of spermatogenic cells increased (P<0.01); Bax and Bcl-w expression of IOD value ratio in spermatogenic cells decreased (P<0.01). There was no significant difference among the high,medium and low LBP treatment groups(P>0.05).Conclusion:LBP has a better therapeutic effect on mice with oligospermia model. The mechanism may be related to the increase of apoptosis profilin bcl-w expression, and the reduction of bax expression.
