Objective:To investigate the effect of Xuebijing injection at different concentration on the functions of endothelial progenitor cells (EPCs) from human umbilical vein blood incubated by lipopolysaccharide(LPS) in vitro.Methods:EPCs which were bought from affiliated Drum Tower hospital of Nanjing University School of Medicine, and assessed with a laser scanning confocal microscope (LSCM). EPCs were cultivated with standard EGM-2MW fluid(control group)or plus LPS(100ng/ml)(LPS group) or first incubated with LPS for24hrs then added Xuebijing injection of different concentrations (XJB-lgroup) or incubated with LPS and Xuebijing injection of different concentrations for48hrs (XJB-2group).The proliferation and adhesion of EPCs were detected with MTT assay. And the influences of LPS and Xuebijing on EPCs’proliferation and adhesion were evaluated.Results:the proliferation and adhesion of EPCs incubated with LPS were both declined significantly (P=0.000,0.042), Both the proliferation and adhesion of EPCs were improved when Xuebijing injection was added, EPCs’proliferation was improved significantly when incubated with Xuebijing injection compared with LPS group and it was related to the doses of Xuebijing injection, and the adhesion was also improved but it wasn’t related to the doses of Xuebijing injection, and there were no significance in5mg/ml and5mg/ml of XBJ-1group (P=0.057, P=0.274respectively). There were all improved significantly in XBJ-2group (P=0.006, P=0.000, P=0.002respectivly)Conclusions:LPS could cut down the proliferation and adhesion of EPCs in some degree. Xuebijing could improve the declined proliferation and adhesion of EPCs which incubated with LPS. It suggested that Xuebijing injection may have something to do with the protective effect of the EPCs injured by LPS. Early therapy make the treatment of sepsis induced by G-bacteria more effective.
