ObjectiveTo investigate the Specific nucleotide variations in the human papillomavirus (HPV)16E2DNA sequence in Qingdao of shan dong Province. The association between the sequence variant and different cervical lesions was also assessed.Methods[1]A total of196fresh-frozen tissue samples and61cervical-scrapings were collected from women in the Gynecologic Department, Affiliated Hospital of the Medical college of Qingdao University during June2008to December2010. The LCT tissues include51highgrade squamous intraepithelial lesion (HSIL) tissues,88lowgrade squamous intraepithelial lesion (LSIL) tissues and53atypical squamous cell of undetermined significance(ASCUS).61cervical cancer biopsies, with were diagnosed8adenocacinoma of cervix and53squamous cervical cancer by pathology.[2] DNA samples were extracted from the cervical scrapes and tissues.[3] DNA samples were amplified by polymerase chain reaction(PCR), using specific primers MY09/11for HPV infection, specific HPV16primers to identify the positive tissues.[4] DNA samples extracted from the HPV16positive tissues were amplified by the primers for E2.[5] the PCR fragments of HPV16E2gene were sequenced and compared with the type stain by DNASTAR.Result[1] The positive rate of HPV and HPV16was82.9%and51.6%respectively in Qingdao, Shandong.[2]A total of11nucleotide mutations were identified, while10of which resulted in amino acid changes. The hot spots of mutation were nt,3410(63.2%),3159(55.9%),3249(55.9%).The analysis revealed that there was no relation between the E2mutation and degree of dysplasia (P>0.05). The integration ratio was65%(14of40) in the cervical cancer specimens, significantly more frequent than in CIN3lesions (6of43)(P<0.001).ConclusionIn this study, there were several E2sequence variations in HPV-16isolates in qingdao of china, G2828A, T3274G, T3384C, T3524C were found for the first time. These variants may be associated with the developing of cervical cancer.
