Background:CPFPI is traditional Chinese medicine compound which had evidenced to resch good therapeutic effect on stroke by our team. Our preliminary works carried out were the preparation process and pharmacodynamic tests of the preparation on the rat stroke. To develop the CPFPI to be a new drug as a Chinese traditional medicine, this article observed effects of CPFPI in rats with blood stasis syndrome, which is the basic symptom of stroke. This experiment got support by scientific and technological project of Chongqing(CSTC,2010AC5011).Objective:Establishment the model of rats with Qi-deficiency and blood-stasis syndrome, Yin Asthenia and Blood Stasis, Toxic heat and blood stasis syndrome, to investigate the effects of Catalpol and puerarin freeze-dried powder for injection (CPFPI) on the coagulation function, hemorheology and NO in rats with blood stasis syndrome, to explore the effect of CPFPI for blood circulation.Methods:1. Modeling method Qi-deficiency and blood-stasis syndrome modeling method The model of rats with Qi-deficiency and blood-stasis syndrome was estabished by hunger, fatigue, cold wetness, panic and high fat diet, once a day, for7days.Deficiency symptoms, stasis symptoms and elevated hemorheology is standard of the medel success.Yin asthenia and blood stasis syndrome modeling method Intramuscular injection Hydrocotisone10mg-kg-1continuous14days to rats. The14days and15days, before get blood1hour, subcutaneous injection Epinephrine0.5mg-kg-1.Toxic heat and blood stasis syndrome modeling method Except control group, the other group administration7days, then, injection LPS500μg-kg-1to rats, after injection LPS5hours measure rectal temperature,24hours measure index.2. Grouping and administrationQi-deficiency and blood-stasis syndrome grouping and administration The rats were divided into control group, model group, xiangdan group, CPFPI16.40mg·kg-1group, CPFPI32.70mg-kg-1group and CPFPI65.40mg·kg-1group by chance(N=10). After statistical analysis, there is no significant difference between the indicators in each group before treatment. Continuous intravenous injection once a day, for7days(control and model group were injected with saline),100g injection1mL.Yin asthenia and blood stasis syndrome grouping and administration60rats were randomly divided into control group, model group, xiangdan group, CPFPI16.40mg-kg-1group, CPFPI32.70mg-kg-1group and CPFPI65.40mg-kg-1group(N=10). In the seventh day start intravenous injection CPFPI and xiangdan, once a day, for7days.Toxic heat and blood stasis syndrome grouping and administration60mice were measured rectal temperature, continuous3days. According to body temperature, mice were randomly divided into control group, model group, xiangdan group, CPFPI16.40mg·kg-1group, CPFPI32.70mg-kg-1group and CPFPI65.40mg-kg-1group(N=10), once a day, for7days.3. Detection indexCoagulation time(CT)was observed by glass method, and bleeding time (BT) was measured by tail-cutting method. The effects of CPFPI were also evaluated on prothrombin time(PT), activated partial thromboplastin time (APTT) and thrombin time(TT). HCT was measured by electric resistance method, hemorheology indices were observed by auto-hemorheological instrument. The level of NO in blood serum was measured by NO assay kit.Results:1. Compared with model group, CPFPI65.40mg-kg-1group can prolong CT(P<0.01), the extension rate is56.10%; CPFPI32.70mg-kg-1group and65.40mg-kg-1can prolong BT (P<0.05, P<0.01), the extension rate is26.40%,42.96%; CPFPI32.70mg·kg-1group can prolong PT, APTT(P<0.05); CPFPI65.40mg-kg-1group can prolong PT, APTT, TT(P<0.05, P<0.01).Effects of Catalpol and Puerarin Freeze-dried Powder for Injection on hemorheology in rats with Qi-deficiency and blood-stasis syndrome Compared with the control group, the viscosity of whole blood and plasma, erythrocyte aggregation and rigidity index, and reduced viscosity of whole blood is increased significantly in model group(P<0.05,P<0.01), it means that Qi-deficiency and blood-stasis syndrome model was reproduced successfully. Compared with model group, CPFPI32.70mg-kg-1group can reduce the viscosity of whole blood, erythrocyte aggregation and reduced viscosity of whole blood(P<0.05), CPFPI65.40mg·kg-1group can reduce each index in hemorheology(P<0.05, P<0.01).Effects of Catalpol and Puerarin Freeze-dried Powder for Injection on NO in rats with Qi-deficiency and blood-stasis syndrome Compared with the control group, NO in model group is significantly reduced(P<0.01), Compared with model group, CPFPI32.70mg-kg-1,65.40mg-kg-1group can rise NO(P<0.01).2. Effects of Catalpol and Puerarin Freeze-dried Powder for Injection in rats with Yin asthenia and blood stasisEffects of Catalpol and Puerarin Freeze-dried Powder for Injection on coagulability in rats with Yin asthenia and blood stasis Compared with the control group, PT, APTT and TT is significantly shorten in model group(P<0.01). It means that Yin asthenia and blood stasis model was reproduced successfully. Compared with model group, CPFPI32.70mg-kg-1,65.40mg-kg-1group can significantly prolonged PT, APTT and TT(P<0.05, P<0.01).Effects of Catalpol and Puerarin Freeze-dried Powder for Injection on hemorheology in rats with Yin asthenia and blood stasis Compared with the control group, the viscosity of whole blood and plasma, erythrocyte aggregation and rigidity index, and reduced viscosity of whole blood is increased significantly in model group(P<0.05, P<0.01), it means that qi-deficiency and blood-stasis syndrome model was reproduced successfully. Compared with model group, CPFPI32.70mg-kg-1group can significantly reduce the viscosity of whole blood and plasma, blood plasma viscosity and reduced viscosity of whole blood(P<0.05), CPFPI65.40mg-kg-1group can reduce the viscosity of whole blood and plasma, blood plasma viscosity, erythrocyte aggregation index and reduced viscosity of whole blood(P<0.05, P<0.01).Effects of Catalpol and Puerarin Freeze-dried Powder for Injection on NO in rats with Yin asthenia and blood stasis Compared with the control group, NO in model group is significantly reduced(.P<0.01), CPFPI32.70mg·kg-1group and65.40mg·kg-1group can raise the level of NO in blood serum(P<0.01).3. Effects of Catalpol and Puerarin Freeze-dried Powder for Injection in rats with Toxic heat and blood stasis syndromeEffects of Catalpol and Puerarin Freeze-dried Powder for Injection on boby temperature in rats with Toxic heat and blood stasis syndrome Compared with the control group, the body temperature of model group was increased significantly(P<0.01), it means that Toxic heat and blood stasis syndrome model was reproduced successfully. Compared with model group,32.7mg·kg-1and65.40mg·kg-1group can reduce body temperature of fever rats(P<0.01).Effects of Catalpol and Puerarin Freeze-dried Powder for Injection on coagulability in rats with Toxic heat and blood stasis syndrome Compared with the control group, PT, APTT and TT was significantly shorten in model group(P<0.05, P<0.01). It means that Toxic heat and blood stasis syndrome model was reproduced successfully. Compared with model group, CPFPI32.70mg·kg-1group can prolonged TT(P<0.05); CPFPI65.40mg·kg-1group can significantly prolonged PT and TT(P<0.05, P<0.01).Effects of Catalpol and Puerarin Freeze-dried Powder for Injection on hemorheology in rats with Toxic heat and blood stasis syndrome The viscosity of whole blood and plasma, erythrocyte aggregation and rigidity index is increased significantly in model group(P<0.01). It means that Toxic heat and blood stasis syndrome model was reproduced successfully. Compared with model group, CPFPI32.70mg·kg-1group can reduce the viscosity of whole blood (P<0.05), CPFPI65.40mg·kg-1group can reduce the viscosity of whole blood, reduced viscosity of whole blood, erythrocyte aggregation index(P<0.05, P<0.01).Conclusions:CPFPI can improve the abnormal coagulation function, hemorheology and raise the level of NO in rats with Syndrome of blood stasis. It show that CPFPI have the effect of blood circulation. To development CPFPI for blood circulation traditional Chinese drug research provided an experiment basis.
