Objective:to investigate the effects of elemene on gastric cancer cell HGC-27 in vivo and in vitro, and to explore the potential mechanisms.Methods:In in vitro study, gastric cancer cell line HGC-27 were treated with different concentrations of elemene, and dose relationship was determined according to results of cell-IQ; the effect of elemene on cell apoptosis was detected by flow cytometry. In in vivo study, a nude mouse model of peritoneal metastasis was employed and treated with elemene; the peritoneal metastasis of human gastric cancer cell was assessed by peritoneal cancer index (PCI); the expression of Ki67, Bcl-2, CD31, VEGF were detected by immunohistochemistry.Result:Elemene suppressed proliferation of HGC-27 in a dose and time dependent manner, and the highest effect was seen at100μg/ml concentration and at 4-19 h after treatment. Elemene induced cell apoptosis at 24 h after treatment, the apoptotic rate were 49.56±4.33% in elemene treatment group and 0.052±0.02% in control group. Elemene also significantly decreased PCI and down-regulated expression of Ki67 in the nude mouse model of peritoneal metastasis.Conclusion1. Cell-IQ can be used in determination of cellular toxicity, activity and proliferation; 2. Elemene suppresses the toxicity, activity and proliferation of human gastric cancer cell, and most effectively at 100μg/ml concentration and 6-48 h after treatment;3. The suppression of elemene on human gastric cancer cell is related to its induction of apoptosis;4. Elemene for human gastric cancer peritoneal metastasis in nude mice have a clear preventive effect.
