| Objectives:To observe the effect of Zhong feng zao bei hua tan Capsules(ZFZBHTC) on nerve protection and the expression of basic fibroblast growth factor (bFGF) in the rats with cerebral ischemia-reperfusion injury, and further explore the protection and mechanism of ZFZBHTC on the cerebral ischemia-reperfusion injury.Methods:75SD rats were randomly divided into sham operation group, ischemia-reperfusion(I-R) group, ZFZBHTC high-dose group and ZFZBHTC middle-dose groupand ZFZBHTC low-dose group,15rats in each group. The rat model of middle cerebral Ischemia(2h)-reperfusion(24h) was prepared by Zea Longa method. Then, each group was observed about behavior changes, cerebral infarction area, line hematoxylin-eosin staining of ischemic region of pathological changes, applying Immunohistochemistry to assay bFGF expression.Results:1Neurological assessment Sham operation group rats have no neurological impairment; I-R group rats have clear neurological impairment; and compared to I-R group, the ZFZBHTC group rats have significantly improved neurological function, and the ZFZBHTC high dose group rats have improved neurological function in the most obvious(P<0.01).2measurement of cerebral infarction Sham operation group rats have no infarction; and compared to I-R group rats, the cerebral infarction areas of ZFZBHTC high, middle, or low dose group rats have significantly reduced, the differences are statistical, and the reduction of the cerebral infarction areas of ZFZBHTC high-dose group rats is the most obvious(P<0.01); but compared ZFZBHTC middle-dose group rats to ZFZBHTC low-dose group rats, the differences have no statistics.3Expression of bFGF After the Cerebral ischemia-reperfusion, Compared with sham operation group, I-R group presented the increase of bFGF expression in cortex and hippocampus. Compared with I-R group, bFGF expression were further increased distinctly in ZFZBHTC groups, of which, the ZFZBHTC high-dose group presented the most significant effect on nerve protection, integrated optical density of positive cells in areas of bFGF is (153.30±3.77). Compared to the ZFZBHTC middle and low dose group, the ZFZBHTC high-dose group improvement is the most obvious, and differences are statistically (P<0.01).4ã€pathological variation The brain tissue structure of Sham operation group rats have not seen infarct formation. But the brain tissue structure of I-R group rats presented neurons and glial cells swelling, arrange disorder, clear infarction, reduction in the number of neurons; and pipe week height edema, structural ambiguities, clearly inflammatory cell infiltration. The brain tissue structure of ZFZBHTC high, middle, and low dose group rats have seen different degrees of neurons and glial cells arranged in mild disorder, part of the cell nucleus appears contracted, deep dyed and swelling; vascular mild edema, part of the expansion of the vascular compression deformation, pipe week inflammatory cell infiltration, intercellular mild edema. Comparison with I-R group, the pathological damage of ZFZBHTC high, middle, and low dose group is all reduced; and comparison with ZFZBHTC medium and low dose group, the pathological damage of ZFZBHTC high dose group is reduced; and comparison with ZFZBHTC low dose group, the pathological damage of ZFZBHTC medium dose group is reduced.Conclusion:Zhong feng zao bei hua tan Capsules(ZFZBHTC) can improve cerebral ischemia-induced neural behavior disorder, reduce brain infarction, inhibit inflammatory cell infiltration, and increase the bFGF expression of the cerebral ischemia-reperfusion injury to play on the neuroprotective effect. |
