Dregea volubilis, a commonly used Chinese herbal medicine, refers to the dry rhizome of evergreen vine plant, with effects of relieving pain arid anti-cancer. DVE, as an impotant part, was extracted from Dregea volubilis by70%ethanol. Eealy studies found that DVE had some immunosuppressive activity in vivo and in vivo. As a mixture, the compositiosn and structures were unclear. For this reason, we further separated and purificated the monomer ingredients from DVE crude extracts, followed by pharmacological activity detection.In the first section of chapter one, we tentatively identified DVE had immunosuppressive effects of the active site. Experimental results showed that the pharmacological activity of the ethyl acetate layer was best, expecially with the highest concentration in DVE. In the second section of chapter one, the ethyl acetate extract was divided into different components. Monomer components were further isolated and purified. Seven compounds were extracted, and two structures of them were first extracted from Dregea volubilis. Finally, we picked up Compound4, with the best pharmacological activity to do the further research.Compound4could selectively inhibited proliferation of activated T cells, but had no effect on non-activated T cells.In the third section of chapter one, we studied the immunosuppressive activity of Compound4in vitro, and found Compound4to inhibit CD3/CD28-induced T cell proliferation with low cytotoxicity. Compound4didn’t affect the activation process ol T cells, but could promote apoptosis of activated T cells in a dose-dependen1manner. Further research found that Compound4could promote the actived T cells to apoptosis, cause mitochondria) membrane potential collapse, and increase the activities of Caspase-3and Caspase-9, while has no effects on FasL expression and caspase-8activity. Compound4promtes actived T cells apoptosis through the mitochondrial pathway and has no effect on non-actiVed T cells, which manifest that Compound4has high selection and low adverse effects.Lindera aggregata (Sims) Kosterm., the leaf ofplate, belongs to Lindera. There were lots of pharmacological experiments done on its dry tuber. While,little research has been done on the leaves of this plant. Total flavonoids (LA-flavonoids) were extracted from the leaves of Lindera aggregata. In chapter two, I studied the function of LA-flavonoids from the leaf of Lindera aggregate on mice hyperlipidemic syndrome.The mice model with hyperlipidemia was established by feeding with high-fat diet; LA-flavonoids (50,100,200nig/kg) were orally administered daily for4weeks.Then biochemical indexes were detected. In mice with high fat-diet, LA-flavonoids at100and200mg/kg decreased serum TC、TG、LDL-C levels. In addition, LA-flavonoids decreased TC and TG in liver, lipid index, AI and body weights in a dose-dependent fashion. HE staining showed that the lipid drops in hepatocytes were remarkably reduced by LA-flavonoids.At the same time, Comp0und4could increase SOD, GSH and decreased MDA in liver.
