Abstract/Summary:
Objective:To observe the inhibitory effects of matrine and matrine plus cisplatin on transplanted cervical cancer of mice,and analyse if matrine can reduce the side-effect of the treatment with cisplatin on cervical cancer,to discuss the clinical application value and rational solutions of matrine in cervical cancer treatment.Methods:To create U14tumor model and randomly divided into8groups(10mice each)which included negative control group,cisplatin group,high dose group of matrine,medium dose group of matrine,low dose group of matrine and the aforementioned three dose groups of matrine plus cisplatin. The inhibitory effects of these groups of drugs on tumor growth were evaluated by measuring the tumor weight,the reduced rate of tumor growth and the index of thymus and spleen after the treatment;By using Cr,BUN and the general condition of mice of combination groups as index which was in order to observe if drug combination can reduce the side-effect of cisplatin treatment.Results:(1) The tumor weight of experimental groups were less than that in negative control group, the tumor weight was lower in medium dose group of matrine(P<0.05) and statistically significant lower in cisplatin group,high dose group of matrine and three combined treatment groups(P<0.01) that compared with the tumor weight of negative control group; the reduced rate of tumor growth of high, medium dose groups of matrine and combination groups were greater than30%, accounting for the inhibitory effects of matrine to U14solid tumor.Compared to the negative control group and the groups with cisplatin,three dose of matrine groups were all able to improve the thymus and spleen index;(2) The value of Cr and BUN in high dose group of matrine plus cisplatin was significantly lower than that was in cisplatin group(P<0.01),and its difference had statistical significance in medium dose group of matrine plus cisplatin(P<0.05),the general condition of these groups above were better than what was in cisplatin group.Conclusion:Matrine of appropriate dosage has anti-tumor effects to cervical cancer, if combined with cisplatin,matrine will have not only the antitumous effect but also the capability of reducing the toxicity of ciaplatin treatment.In other words,matrine could enhance the inhibiting effect of cisplatin on mice cervical cancer.This provides us theoretical basis for the combination chemotherapy between matrine and cisplatin in the patients of recurrent cervical carcinoma or advanced stage or being too old to tolerate surgery.
