Objective: To study the influence of zidovudine(AZT) to osteosarcomacell line (HOS) in vitro in cell morphology, survival rate and telomeraseactivity. and explore the application prospect of telomerase inhibitors in thetreatment of osteosarcoma.Methods: osteosarcoma cell (HOS) were divide into no drug and AZTgroup, and were named as control group,AZT group.We observed themorphological change of osteosarcoma cells with light microscope; studied thesurvival rate by MTT assay; detected changes of telomerase activity byTRAP-PCR-ELISA.Results: After the effects of AZT, osteosarcoma cell line (HOS) hasundergone an obvious change in shape by size decrease, shrinkage. The shortspindle became polygonal or irregular in shape, and with neighboring cellsdetachment. Compared with control group, the survial rates in AZT groupswere significantly decreased (p<0.05). in a time-and dose-dependentmanner.Its IC50was100μmolL-1. The telomerase activity of Hos cells wasdecreased by55.74%after treated with100μmolL-1AZT for72h.Conclusion: AZT has an explicit of inhibiting proliferation of HOS cells,and AZT can reduce the telomerase activity of osteosarcoma cells...
