Objctive:To explore the expressions o f EPO-R,JAK2and STAT-5in the humanbrain with refractory epilepsy and the role in neural apotosis.Background:Experimental modeling and clinical neuroimaging of patients has shownthat recurrent seizures Could induce neuronal apotosis.such brain injury maycontribute to epileptogenesis,impaired cognitive function, so is considered oneof the most appealing candidates for therapeutic approaches toepileptogenesis,experimental study results showed that Epo can decreaseneuronal cell death and reduce the risk of Spontaneous recurrent seizuresdevelopment, The neuroprotection effects of EPO may have relationship withthe pathway of EPO-R/JAK2/STAT-5,some Experimental modeling ofepilepsy have found that the expressions o f EPO-R dramatically elevatedafter SE,But until now,the report about the expression of EPO-R,JAK2andSTAT-5in the human brain with refractory epilepsy have not been seen.Materials and methods:Collecting the informations of24patients (as experimental group) withintractable epilepsy who were in hospital and undergo March2010to July2011,according to relevant stipulations of law,6cases of normal brain tissue (ascontrol group) were obtained from immediately conducted autopsies who deathduo to accidental death or unnatural death during the same term. Gathering thebrain tissues to observe the above EPO-R、JAK2and STAT-5expression of thebrain tissue through immunohistochemical, then statistical analysis of the result row.Result:1.EPO-R、JAK2'STAT-5are expressed in both experimental and controlgroups. In experimental group, the positive-cell number were41.05±2.40、50.21±2.50、60.18±2.84. under light microscope (400×power). While in controlgroup, the positive-cell number were23.00±0.49、27.00±0.88、25.93±0.33.There were significant differences between the2groups(P<0.001).2.There were the pathologic and ultrastructural changes of the humanbrain with refractory epilepsy. in the optical microscope we can observe thatthe distribution of neurons was uneven,immature neurons were visible;Thenuclei was vacuolated and the cytoplasm was less,We can see hyalomitomeacidophilic body,The neurons became triangular due to degeneration.Theproliferation and hyperemia appeared in small vascular and glial cells,In thetransmission electron microscope we observed degeneration and necrosis of thenerve cells,The nuclear became pyknotic, karyolemma were breakage and evendissolved,Nucleolis were dyssymmetry; The mitochondria and astrocytes wereswelling,We also can see part of the mitochondrial cristae was abnormal.Conclusion:1.we find neuronal apotosis in the human brain with refractory epilepsy.2.Intractable epilepsy betrayed higher EPO-R、 JAK2andSTAT-5expression in comparison to control group on LM and EM. the highexpression of EPO-R、JAK2andSTAT-5is unrelated with course andfrequency of epileptic seizures.3.the pathway of EPO-R/JAK2/STAT-5may be involved in thepathophysiological processes of neural protective effect of endogenous EPOagainst brain injury induced by epileptic seizures.
