Objective:Explore the changes of rat peripheral blood, spleen and bone marrow myeloid inhibit cells,(MDSCs) under the different fluid resuscitation in hemorrhagic shock.Methods:Hemorrhagic shock rat model is made by controlled bleed through carotid artery. Then during the early phase of fluid resuscitation, we use normal saline, hypertonic saline and hydroxyethyl starch, followed by blood reinfusion phase,and put to death after three hours. After we executed the rat, we should gain rat peripheral blood, spleen and bone marrow cell suspension quickly. And the rats are divided into five groups:control group, sham group, normal saline group, hypertonic saline group, hydroxyethyl starch group. Get the number of myeloid suppressor cells in peripheral blood, spleen and bone marrow, using the flow cytometry observation.Results:Compared with control group, the ratio of MDSCs in peripheral blood and bone marrow, is changed obviously in sham group, saline group, hypertonic saline group, hydroxyethyl starch group, and it showed statistically significant (P<0.05), while the ratio of MDSCs in spleen showed no statistically significant (P=0.282). Compared with sham group, the ratio of MDSCs in peripheral blood and spleen, showed no statistically significance in saline group and hydroxyethyl starch group (P>0.05), but the ratio of MDSCs in peripheral blood increased, compared with the sham group, and that in bone marrow reduced. Besides, there’s no significance difference between saline group and hydroxyethyl starch group. The containing of MDSCs in the peripheral blood of hypertonic saline group was remarkably higher than sham group, saline group and hydroxyethyl starch group, and decrease in bone marrow, and it showed statistical significance (P<0.05).Conclusions:From the experiment results, we can speculate:control with normal saline and hydroxyethyl starch, hypertonic sodium chloride solution may be better to promote the generation of MDSCs in the peripheral blood, so that MDSCs can inhibit immunoreaction during hemorrhagic shock.
