Background and Purpose:Atherosclerosis (AS) is a pathological and pathophysiological basis of the ischemicheart and cerebrovascular diseases, which has a relationship with a variety of diseases, is oneof the most common diseases in the elderly. The Pathological changes caused bycardiovascular and cerebral vascular damage leading to acute coronary syndrome and stroke,has become the most serious disease with the highest mortality, morbidity and disability rateworldwide, far more threatening than malignant. A lot of basic experimental and clinicalobservation committed to research on AS pathogenesis and the method of treatment in orderto fundamentally solve AS. Therefore, to explore the molecular mechanisms of theatherosclerosis process, the effect of drugs on the vascular endothelial inflammatoryresponse may be a new target for anti-atherosclerosis treatment. From his study, to establishmice AS model through the high-fat diet, we can understand the expressions of PAI-1å'ŒTNF-α in local Pathological changes of mice,and observe the effect of resveratrol on theimpact of PAI-1and TNF-α and its resistance to the mechanism of AS.Methods:8week old male ApoE-/-mice16,weight18to22g,were purchased from ExperimentalAnimal Center of Peking University,and randomly divided into a normal diet group(8):normal diet+salineï¼› b cholesterol diet group(8):high-fat diet+saline;c resveratrolgroup(8):feeding high-fat diet+resveratrol.They were given the diet for12weeks,cervicalamputation sudden death in mice,removing from the aortic root at the aortic arch0.5cm,placed in4%formaldehyde solution preserved,paraffin embedded,usingimmunohistochemical methods, measurement and arterial plaque plasminogen activatorinhibitor type-1(PAI-1), tumor necrosis factor-α(TNF-α).,measured by the application ofstatistical software SPSS12.0gray value differences between the groups compared.Results:Immunohistochemical analysis showed that we can and seen a lot of brown-yellowmembrane staining granular substance had been seen in the intimal thickening in cholesteroldiet group,partial integration into the film especially in endometrial. However staining substance in the resveratrol group was groups were significantly different significantlydifferent(P<0.01).Conclusion:1.The Percentage of the positive stained of PAI-1and TNF-α is much higher in the miceof AS.PAI-1and TNF-α over-expression may be one of mechanisms of AS.2. Resveratrolinhibits the percentage of positive stained of PAI-1and TNF-α may be one mechanism ofAS.3. Measuring PAI-1and TNF-α has great significance for early diagnosis of AS diseaseand monitoring of AS’occurrence,development and prognosis.
