Objective: To establish lithium-pilocarpie model of temporal lobe epilepsyin in the acute phase test, and make use of the Ethosuximide、Lamotrigine to intervene, in order to find out antiepileptic effect of chronic spontaneous period and neuroprotective effect.Methods (1)Healthy adult female wister rats were intraperitoneal injection of lithium-pilocarpine induced status epilepticus(SE)by according to the classification of the IV Racine method above level. SE last60minutes after the injection of diazepam terminate attack, live be regarded as successful made model. Normal control group with the same volume of0.9%sodium chloride replace of lithium-pilocarpine and diazepam. successful model of the rats were randomly divided in to5groups:model control group, ESM low-dose (25mg/kg,2times/d) treatment group, ESM high-dose (50mg/kg,2times/d) treatment group,LTG low-dose (25mg/kg,2times/d) treatment group and LTG high-dose (50mg/kg,2times/d) treatment group, each group of only8. Drug intervention group, respectively in the24hours after the end of the SE to medicine, and continuous to medicine ten days.(2) Observation experimental process of seizures grade and attack frequency rat seizures, explain the differences of data among groups.(3) Nissl and glial fibrillary acidic protein Immunohistochemistry to observes, the hippocampus were observed CA3, CA1area if there is situation appear of neuron loss, astrocytes hyperplasia, analyzes each group have significant differences between indexResults:Compared with the control group, weight, appetite, squares crossed, rear times and grooming in open-field test in the model group decreased remarkably, while epilepsy seizure intensity, frequency, time of staying in central square and defecation increased significantly(P <0.05). And in the other3groups, compared with the model group, weight, appetite, squares crossed, rear times and grooming in open-field test increased remarkably, while epilepsy seizure intensity, frequency, time of staying in central square and defecation decreased significantly(P <0.05). The LTG high dose group, compared with the group and low dose group, also produced the same results (P<0.05), which had statistical significance. However, the results of the group and low dose group were different, neither of which existed statistical significance. showed accumulated spike waves and short-time latency during acute stage, but epileptic waves and long-time latency during the chronic phase. Compared with the control group, latency in the other.Conclusions:(1) established in this study lithium-pilocarpine seizure pattern is very close to the human characteristics of chronic temporal lobe epilepsy, spontaneous recurrent epileptic seizures in human chronic depression, can simulate the human state of with epilepsy is associated in epilepsy research for an ideal animal model.(2) LTG inhibited the epileptic discharge, significantly reduced the number of spontaneous seizures, reducing neuronal damage and glial cell proliferation, has antiepileptic effects (3) ESM has no effect on eitherantiepileptic or neuroprotection, in lithium-pilocarpie model of emporal lobe epilepsyin model.
