Objective To investigate the effect of sufentanil precondition(SPC) and opioid receptoron renal injury after intestinal ischemia reperfusion(IIR).Methods A total of72adult and health SD rats, female or male, weighting200-250gwere randomly divided into nine group(n=8):①sham operation group(S): performed by thesame way of intestinal ischemia reperfusion group except for ligating superiormesenteric artery;②intestinal ischemia reperfusion group(IIR): ligating superiormesenteric artery for1hours,then reperfusion for6hours;③sufentanil precondition group(SPC): receiving sufentanilinfusion at the dose of10μg/kg ten minutes before IIR;④CTOP interventiongroup(CTOP+SPC): receiving CTOP infusion at the dose of1mg/kg10minutes beforesufentanil precondition;⑤NTD intervention group(NTD+SPC): receiving NTD infusion at thedose of10mg/kg10minutes before sufentanil precondition;⑥nor-BNI interventiongroup(nor-BNI+SPC): receiving nor-BNI infusion at the dose of10mg/kg10minutes beforesufentanil precondition;⑦CTOP group(CTOP): receiving CTOP infusion at the dose of1mg/kg10minutes before IIR;⑧NTD group(NTD): receiving NTD infusion at the dose of10mg/kg10minutes before IIR;⑨receiving nor-BNI infusion at the dose of10mg/kg10minutes before IIR. The animals of S group were killed at6h after operation and the animals ofother groups were killed at6h after IIR and the right kidney of the rats were collected. Lightmicroscopy was performed to verify the morphologic changes of renal tissue. The expression ofBcl-2and Bax protein in renal tissue were detected with immunohistochemistry and MIAS. Theapoptosis index(AI) was measured by TUNEL method.Results①Compared with S group, the expressions of Bc1-2and Bax increased whilethe radio of Bc1-2to Bax decreased and the AI increased(P<0.05) in IIR group;②Comparedwith IIR group, the expression of Bc1-2increased, the expression of Bax decreased, the radio ofBal-2to Bax increased and the AI decreased(P<0.05) in SPC, CTOP+SPC, NTD+SPC andnor-BNI+SPC groups;③Compared with SPC and CTOP+SPC groups, the expression ofBc1-2decreased, the expression of Bax increased, the radio of Bc1-2to Bax decreased and theAI increased(P<0.05) in NTD+SPC and nor-BNI+SPC groups;④There weren’t muchdifferences between IIR group, CTOP group, NTD group and nor-BNI group in the expressionsof Bc1-2and Bax, the radio of Bc1-2to Bax and the AI.Conclusions①Sufentanil preconditioning can significantly decrease the renal injury ofrats after intestinal ischemia reperfusion.②Sufentanil preconditioning produce a protective effect through δ-opioid receptor and κ-opioid receptor mediate on renal injury after intestinalischemia reperfusion, and it could not produce an effect on renal injury after intestinal ischemiareperfusion just by μ, δ, κ-opioid receptor antagonist solely.③The mechanism of protectionmay be related to induction of Bcl-2protein expression and inhibition of Bax protein expressionby sunfentanil preconditioning.
