ObjectiveThis is a research to investigate the expression trends of Vimentin and α-SMA in renal tissuefrom unilateral ureteral obstruction(UUO)rats and the relationship of Vimentin,α-SMA andrenal interstitial fibrosis,meanwhile,to study the intervention effects of Spirolactone.MethodsThe UUO method were used to establish the models.A total of96young Wistar male rats wererandomly divided into Sham-operated group,UUO model group and Spirolactone-treated group.At the days7,14,21and28after operation,every8rats from each group were sacrificed fordynamic evaluated the degree of renal interstitial injury of the three groups by Masson stainingin the left kidney tissue,and SABC immunohistochemical analysis was used to determine theexpression levels of Vimentin and α-SMA.Results(1)Morphological changes:At the day7after operation,renal interstitial was edematous and widen,part of the renal tubule dilated and slight inflammatory cells infiltrated inUUO group;at the day14renal tubule dilated and inflammatory cells infiltrated into renal tissue;It was showed obviously at the day21that there were more and more inflammatory cells infiltrated diffusely,part of epithelial cells fell off and atrophied;at day28renal tubule structure was damaged seriously and atrophied more and more.The renal interstitial fibrosis were diffusely.There were no obviously change of renal organization of Sham-operated group rats.Compared with UUO model group,renalinterstitial damage index were significantly reduced in different time points in Spirolactone-treated group(P<0.01),but still higher than Sham-operated group(P<0.01).It was obviously that the dilation and atrophy of renal tubule were improved and the extent of tubulointerstitial fibrosis was reduced.(2)Expression of Vimentin and α-SMA of immunohistochemical method detection:①Sham-operated group:Vimentin was expressed slightly in renal tubule and fibroblasts,andalmost no exist in glomerular and vascular.α-SMA existed in the blood vessels onlya few.②UUO group:at the day7after operation,renal tubule epithelial cells expressed a few Vimentin,and the level of expression of vimentin was increased gradually.There were lots of vimentin expressed in renal tubule epithelial cells and interstitial atday21,28,but the level of expression of glomerular was not increasing obviously;the expression of α-SMA was intensivly increased with the extension of the durationof the experiment in renal tubules and interstitial.At the day7,14,21and28,the levelof expression of α-SMA was increase progressively,and higher than Spirolactone-treated group significantly(P<0.01).At each time point the difference in UUO groups was significant(P<0.01).③Spirolactone-treated group:the parts of expression of vimentin and α-SMA were the same as UUO group,but lower than UUO group.(P<0.01).(3)Correlation Analysis:The expression of vimentin and α-SMA were positively correlated with renal fibrosis degree(r=0.647,P<0.01)of UUO group.But the rest of the two groups have no correlation.ConclusionIn the course of tubulointerstitial lesions with young rats,the up-regulated expression ofvimentin and α-SMAhave important pathogenic significance.In he UUO method,vimentin andα-SMA were positively correlated,it was signified that they have function of acceleratingtubulointerstitial fibrosis.And spironolactone plays an important role in relieving renal fibrosisby suppressing the expression of vimentin and α-SMA.
