Abstract/Summary:
Aim:To investigate the feasibility and efficiency of killing human osteosarcoma cells mg-63by TK/GCV combined with cDDP.Methods:Restructure and transfect the plasmid pIRES-EGFP-tk to human osteosarcoma cell mg-63.Analyse and observe features of growth of the cell by flow cytometry and inverted fluorescence microscopy. Add GCV and cDDP, study the efficiency of killing the cells mg-63by TK/GCV combined with cDDP.Results:Plasmid pIRES-EGFP-tk was successfully restructured and tk gene was transfected into cells mg-63.It showed green fluorescence from mg-63(tk+) by observing the cells under fluorescence microscope.The rate of transfection detected by flow cytometry is67.7%. The growth of cells mg-63(tk+) was greatly inhibited by GCV administration with10μg/ml and cDDP killed cells mg-63(tk+) at concentration of1μg/ml as lowest concentration.When combined with GCV at low concentration of5μg/ml,the concentration of cDDP can be decreased to0.25μg/ml.Conclusion:TK/GCV mediated by pIRES-EGFP-tk can kill human osteosarcoma cell mg-63. It can enhance the killing efficiency when combined with cDDP.
