Objective:To investigate mutations of oncogene K-ras in colorectal cancer tissues andthe relationship between mutations of K-ras and clinicopathologicalcharacteristics in colorectal cancer.Methods: specimens of 90 patients with colorectal cancer in our department wascollected from All the specimens are DNA sequenced by the sanger methodin order to detect k-ras gene mutation type and mutation location. Then theassociations between K-ras mutation status and clinicopathologicalcharacteristics in colorectal cancer were analyzed.Result:31 cases were detected mutation in 90 cases of colorectal cancer specimens.The mutation rate was 34.4℅. There were 22 cases of mutation occurred incodon 12, 9 cases occurred in codon 13. One case had double mutation atcodon 12.There were no significant differences in age,gender,tumorsize,Dukes stage, hepatic metastases, and was correlated with location oftumor. The frequency of 12∕13 codon mutation is no correlated with age,gender,tumor size,Dukes stage, hepatic metastases, location of tumor.Conclusion1.K-ras gene total mutation rate of 12∕13 codon is 34.4℅in colorectal cancer .2.The frequency of K-ras mutation was correlated with location of tumor incolorectal cancer.3.The frequency of K-ras mutation is no correlated with age, gender ,tumorsize ,Dukes stage, hepatic metastases in CRC.4.The frequency of 12codon single mutation is no correlated with age, gender,tumor size,Dukes stage, location of tumor ,hepatic metastases in CRC.5 The frequency of 13 codon mutation is no correlated with age, gender,tumorsize,Dukes stage, location of tumor ,hepatic metastases in CRC.
