Objective Human Glomerular Mesangial Cells (HMCs) cultured in vitro. HMCs establishing through continued effect of high concentration glucose on cell model of oxidative stress, Exogenous joined adiponectin(ADPN) observation on the proliferation of HMCs under high glucose environment effects of oxidative stress and cell level, Thus revealing ADPN in diabetic nephropathy on HMCs in the process of being to the protection mechanism.Methods①HMCs were cultured with5.5mmol/L glucose (control group),30.0mmol/L glucose (high glucose group),30.0mmol/L glucose and concentration of2.5μg/ml ADPN (ADPN group), respectively. According to the experimental group and culture for24h,48h,72respectivly, the proliferation activity of HMCs was observed by MTT in all time points.②HMCs were cultured once more with5.5mmol/L glucose (control group),30.0mmol/L glucose (high glucose group),30.0mmol/L glucose and concentration of2.5μg/ml ADPN (ADPN group), respectively. After culture for24h,48h,72h, Different points in each group the content of MDA in the cell supernatant was measured by Thibabituric acid (TBA). Different points in each group the vitality of the SOD in the cell supernatant was measured by the xanthineoxidase method, the expression of HO-1mRNA was observed by RT-PCR.The above indexes used for investigating and estimating the effect of HMCs on the influence of oxidative stress.Results Compared with normal group, HMCs proliferation activity has no difference in high glucose group in24h (P>0.05), and that was inhibited in48h、72h(P<0.05), but the proliferation of HMCs was increased apparently after ADPN being added(P<0.05). Meanwhile, the content of SOD and the level of mRNA of HO-1decreased and the content of MDA increased in the high glucose group in the24h、48h、72h compared with normal group (P<0.05). After adding ADPN, which can reduce oxidative stress and increase the antioxidant capacity,the content of SOD、MDA and the level of mRNA of HO-1had being changed, there has statistical difference (P<0.05).Conclusion Long-term high glucose stimulating could inhibit cell proliferation of HMCs, but ADPN could promote the proliferation of the HMCs and protect them in D-glucose of high concentration. The high glucose could enhance the oxidative stress level, but ADPN could chang it. ADPN could protect HMCs in high density glucose.which provided a new forecast indicator and therapeutic target for the diabetic nephropathy.
