BACKGROUND&OBJACTIVE:Nasal polyps is a kind of common nasal clinical diseases. Nasal polyps (NP) are benign lesions arising from the mucosa of the nasal sinuses(commonly at the outflow tract of one or more of the sinuses) or from the mucosa of the nasal cavity. Their etiology remains unclear, but they are known to have associations with allergy, asthma, infection,cystic fibrosis, and aspirin sensitivity.Studies suggest many factors have effect in the formation of Nasal Polyps. Recent studies tend to pay close attention to many kinds of cell factors’role in the process of nasal polyps. In nearest studies, researchers think the process of nasal polyps is the result of imbalance between cell proliferation and apoptosis, many research literature on nasal polyps have reported the process of abnormal cell apoptosis, says many kinds of cell factors’ abnormal expression related with it. The aim of this study is to detect the expression of hypoxia inducible factor-1α(HIF-1α),signal transducers and activators of transcription3(STAT3) and Survivin in nasal polyps and the correlation among them, and to explore the role of them in the pathogenesis of nasal polyps.METHODS:25cases of nasal polyps and15cases of normal inferior turbinate mucosa were enrolled in this study. HIF-1α,STAT3and Survivin in nasal polyps tissues from25 nasal polyposis and15samples of inferior turbinate tissues were detected by immunohistochemistry method.We use SPSS17.0software to analyse all data.p<0.01was considered as statistical significance.RESULTS:Under optical microscope, cytoplasma within the HE-stained nasal polyps tissue is displayed in light pink color, while the nucleus are dark blue or black. The over-dropsical loose connective tissue, which is contained in the nasal polyps tissue, is found in bubble-shaped dilation, accompanied by massive proliferation of beaker cells and gland tissue. Plenty of inflammatory cells are found among cellular matrix, for instance, eosinophile granulocyte, mastocyte, especially the former one. There is new born blood vessel, but no neuronal tissue, in nasal polyps, the basement membrane is usually thickened. On the other hand, there is no beaker cell or over proliferated tissue detected in relative concha nasalis inferior tissue. The crypt is usually in round shape. In contrary to nasal polyps, inflammatory cells are rare, and the number of eosinophile granulocyte is quite limited.HIF-1α-positive signals are mainly located in cytoplasma or nucleus colored as chino. Cells with positive signals are mostly detected in nasal polyps mucous epithelium, gland, and blood vessel endothelium. There is also HIF-1α-positive signals in cellular matrix. However, majority of STAT3positive signals are distributed in cellular matrix colored as chino or brown. Minority of them are found in nucleus. In staining test they are often chino or brown. Surviving is mostly expressed in cytoplasma among nasal polyps, other than in nucleus. Surviving is mainly located in the epithelia cells on the mucous membrane, endothelial cells in the gland underneath mucous membrane of nasal polyps tissue, and in inflammatory cells. Similarly, they are colored in chino or brown.The positive rates of HIF-1α,STAT3and Survivin expression in nasal polyps were76%,68%and64%respectively, all were significantly higher than those in control group(P<0.01).In nasal polyp tissues, the expression degree of HIF-1α,STAT3,and Survivin display positive correlation between each of them(P<0.01).CONCLUSION:HIF-la,STAT3, and Survivin may play an important role in the pathogenetic mechanism of nasal polyps. And the significant positive correlations among them in the tissues of nasal polyps, suggesting that the roles of HIF-1α,STAT3and Survivin are not isolated, but may have collaborative effect.
