Objective To observe the inhibition in vitro of Polysaccharide fromSugarcane Leaves(GZY)to NPC Cell line Cne2and discuss its mechanism.Methods GZY were extracted by hot-water extraction.And GZY werepurified by papain enzymic hydrolysis method and decolouring by H2O2. GZYwere further purified by DEAE-cellulose (OH-) ion–exchange. The anti-tumoractivity of GZY in vitro was analyzed by MTT. The morphological changes ofapoptotic cells were evaluated by transmission electronic microscopy. Cellsapoptosis were measured by Flow cytometry. The expression levels of Bax andBcl-2were detected by real-time PCR.Results Inhibitions and apoptosis on Cne2cells by GZY were significantlyincreased and in concentration-dependent (P <0.01). Apoptotic body could be found in Cne2cells induced by conditioned medium at160ug/ml by GZY.Withconcentration increase the expression of Bax in Cne2cells increased and Bcl-2decreased.Conclusion GZY has obvious inhibition on NPC Cell Cne2,would induceapoptosis to inhibite the growth of Cne2cells.The activation of bax and thesuppression of bcl-2may contribute to the apoptosis mechanism....
