Objective:To investigate association between the polymorphisms of Apolipoprotein C-Ⅲ(ApoC-Ⅲ), Apolipoprotein E(ApoE) gene and human longevity in Bama County, and explore the interaction between the environmental factors of the longevity district and the genetic polymorphisms of these two genes.Methods:Study was conducted in Bama County of Guangxi Zhuang Autonomous Region. The longevity district of Pingdong Xishan Jiazhuan was measured by the population distribution of longevity to be the top three in Bama(105/105、80/105、76/105), and the non-longevity district of Nashe Jusang was measured by that to be the last or last(21/105、15/105). Collected the external control of Lihu and Xiaochang townships of Nandan County as these two places are similar in natural environment and habits. Here we collected152longevity persons aged90or more from longevity district of Bama County as the longevous group. The controls were composed of3groups. The control1was from longevity district, the control2was from no-longevity district, the control3was from no-longevity of Nandan County. The control1was composed of342aged people between22and89years who had no-longevity families. The control2was composed of308aged people between22and89yeas who had no-longevity families. The control3was composed of240aged people between22and89years who had no-longevity families. Then combined the longevous group and the control1,2as the Bama group. The longevous group and the Bama group were the experiment groups. After getting subjects’permission, the blood samples were collected and DNA was extracted from them, then detecting the polymorphisms of ApoC-Ⅲ(2542052), ApoE(7412/429358) gene with Taqman SNP Genotyping Assays. All data were performed with x2statistics and logistic regression analysis.Results:1. No statistically significant from Hardy-Weinberg Equilibrium (HWE) for the polymorphisms in rs2542052/7412/429358genes for the study population were observed(P>0.05).2. Comparison of the distribution of ApoC-Ⅲ(2542052) between longevous group and control1,3, Bama group and control3:There were significant differences in the distributions of genotype between these groups; Comparison of the distribution of ApoE(429358) between Bama group and control3:There were significant differences in the distributions of genotype between the two groups.3.The results of the experiment groups compared with each control groups, the genetic polymorphism associating with Bama longevity were the followings. Between longevous group and control1, compared with the individual harboring genotypes of rs2542052, the genotype was no associated with the Bama longevity; Between longevous group and control2, the AA and AC+AA genotypes of rs2542052might ben the risk factors of the Bama longevity(OR=2.553,95%CI:1.325-4.921, OR=1.976,95%CI:1.135-3.44); Between longevous group and control3, the AC/AA/AC+AA genotypes of rs2542052might ben the risk factors of the Bama longevity(OR=2.001,95%CI:1.271-3.149, OR=1.595%CI:1.009-2.323); The other genotypes of rs7412/429358were no associated with the Bama longevity.4. The longevous group compared with the control1and2,3and the Bama group compared with the control3, the distribution of minority and majority allele in the gene locus(2542052) had statistically significant(P<0.05) and in the gene locus(7412/429358) had no statistically significant(P>0.05).5.The interaction analysis showed that between longevous group and control1, the genetic polymorphism of rs2542052had interaction with smoking(P<0.05). Comparing with the CC genotype carriers with no smoking, the AC+AA genotype carriers with smoking was the risk factor of the Bama longevity(OR=3.742,95%CI:1.719-8.143).Between longevous group and control2, the genetic polymorphism of rs2542052had interaction with smoking(P<0.05) and drinking(P<0.05).(1)Comparing with the CC genotype carriers with no smoking, the AC+AA genotype carriers with no or yes smoking were the risk factors of the Bama longevity(OR=3.107,95%CI:1.587-6.08, OR=9.127,95%CI:3.805-21.894).(2) Comparing with the CC genotype carriers with no dinking, the AC+AA genotype carriers with no drinking and the CC/AC+AA genotype carriers with drinking were the risk factors of the Bama longevity(OR=2.448,95%CI:1.248-4.799,OR=4.291,95%CI:1.324-13.909,OR=5.071,95%CI:2.353-10.927).Between longevous group and control3, the genetic polymorphism of rs2542052had interaction with drinking(P<0.05) and no interaction with smoking(P>0.05).(1)Comparing with the CC genotype carriers with no smoking, the AC+AA genotype carriers with smoking was the risk factor fo the Bama longevity(OR=2.269,95%CI:1.252-4.113).(2) Comparing with the CC genotype carriers with no drinking, the CC and the CT+TT genotype carriers with drinking were the risk factors of the Bama longevity(OR=2.634,95%CI:1.339-5.179, OR=6.736,95%CI:2.035-22.297, OR=7.037,95%CI:3.108-15.931).The genetic polymorphism of rs429358had interaction with drinking(P<0.05) and no interaction with smoking(P>0.05). Comparing with TT genotype carriers with no smoking, the TT and CT+CC genotype carriers with drinking were the risk factors of the Bama longevity(OR=4.777,95%CI:2.428-9.398, OR=2.152,95%CI:1.04-4.452).Between Bama group and control3, the genetic polymorphism of rs2542052had interaction with drinking(P<0.05) and no interaction with smoking(P>0.05). Comparing with the CC genotype carriers with no drinking, the AC+AA genotype carriers with drinking was the risk factor of the Bama longevity(OR=2.936,95%CI:1.57-5.49).The genetic polymorphism of rs429358had interaction with drinking(P<0.05) and no interaction with smoking(P>0.05). Comparing with the TT genotype carriers with no smoking, the TT and CT+CC genotype carriers with drinking were the risk factors of the Bama longevity(OR=1.742,95%CI:1.185-2.561, OR=2.526,95%CI:1.518-4.203).6. The MAF in the different ethnic of the same race, the results showed that rs2542052/7412had no statistically significant(P>0.05), rs429358had statistically significant(P<0.05).Conclusions:1. The polymorphism of ApoC-III SNP locus(2542052) might be association with Bama longevity; The polymorphism of ApoE SNP locus (7412) was no association with Bama longevity; The polymorphism of ApoE SNP locus (429358) association with Bama longevity needed further study.2. There might be interaction between the genetic polymorphism of ApoC-III SNP locus(2542052) and the habits(smoking, drinking), also might have association with Bama longevity; There was no interaction between the genetic polymorphism of ApoE SNP locus(7412) and the habit(drinking), the interaction between he genetic polymorphism of ApoE SNP locus(7412) and the habit(smoking) needed further study; There was interaction between the genetic polymorphism of ApoE SNP locus(429358) and the habit(drinking), also might have association with Bama longevity, and no interaction between the genetic polymorphism of ApoE SNP locus(429358) and the habit(smoking).
