Objective:To investigate the effect of oxymatrine-cisplatin and oxymatrine-oxaliplatin on cell proliferation in human hepatoma cell line Bel-7404and its mechanism, Providding the theory basis for the combination of traditional medicine with chemotherapy to cure hepatocarcinoma.Methods:Human hepatocelluar carcinoma Bel-7404cells were cultured in vitro and affected by oxymatrine, cisplatin, oxaliplatin, oxymatrine-cisplatin, oxymatrine-oxaliplatin in different dose and different time respectively. MTT-test was used to estimate the inhibition of cell proliferation, Inverted microscope was employed to observe morphologic changes, flow cytometry was applied to analyze the distribution of cell cycle and cell apoptosis.Results:Oxymatrine, cisplatin and oxaliplatin had obvious inhibiting effect on the proliferation of human hepatoma cell line Bel-7404which depended on exposure time and dose (p<0.05) and oxaliplatin was superior to cisplatin(p<0.05). There were additive effects when combine oxymatrine of2mg/ml with cisplatin of2ug/ml after24h while synergistic effects after48h and72h, There were synergistic effects when combine oxymatrine of2mg/ml with oxaliplatin of2ug/ml after24h,48h and72h, and oxymatrine-oxaliplatin was superior to oxymatrine-cisplatin (p<0.05). Observed by inverted microscope, adhesion and colony formation of cells depressed, cells became much smaller and most of them shaped long and narrow after drug treatment. Flow cytometry analysis showed the proportion of cells increased at G0/G1phase while it decreased at S phase, indicating that drugs blocked cells at G0/G1phase, cell apoptosis rate increased and mainly by early apoptosis constituted, and the effect of combined group significantly excelled the single work.(p<0.05)Conclusions:The results suggest that oxymatrine, cisplatin and oxaliplatin would have inhibiting effect on cell proliferation in human hepatocelluar carcinoma cell line Bel-7404, which would be strengthened and attain synergism when oxymatrine worked combinedly with cisplatin or oxaliplatin and oxaliplatin was superior to cisplatin whether it worked separately or combinedly. Its mechanism may be related to the blocking of cell cycle at G0/G1phase and inducing cell apoptosis.
